The single nucleotide polymorphisms (SNPs) in Apo-Ⅱgene,OVR gene and VTGⅡgene were detected and their association with content of cholesterol in egg yolk was analyzed in Princess Chickens.The polymorphism in exons of Apo-Ⅱgene, OVR gene and VTGⅡgene was amplificating using PCR-SSCP and the associativity between controlling genes and content of cholesterol in egg yolk was approaching, which were in order to providing molecular genetic markers for breeding of low content of cholesterol laying chicken strains.The results showed:1,There were no mutation site in exons 2 and 3 in Apo-Ⅱgene and 1 and 2 in OVR gene.2,There was no mutation site in exons 6, 26, 27, 34 and 35 in VTGⅡgene and there were 4 sites in exons 22 and 33(C13161T,A17698T,T17719G,G17764A). There were 2 genotypes in site C13161T(CC and CT), which was silent mutation. There were 2 genotypes in site A17689T(TT and AT), which existed in intron. There were 3 genotypes in site T17719G(TT,TG,GG), which resulted in amino acid change of tryptophane→glycine. There were 2 genotypes in site G17764A(GG,GA), which resulted in amino acid change of glutamic acid→lysine.3,The test of Hardy-Weinberg equilibrium showed that site C13161T and A17698T were in equilibrium, but site T17719G and G17764A were not in equilibrium. Genetic diversity analysis showed that: the sites C13161T and A17698T were lowly polymorphic, and the sites T17719G and G17764A were moderately polymorphic, in which the site G17764A had the most highly polymorphic, biggest mutation degree and best potentiality. This indicated that the researched chicken groups had a rich genetic diversity.4,Statistical analysis showed that there had no significant differences(P > 0.05) between the different genotypes in exon26 and exon33。...
