The single nucleotide polymorphisms(SNPs) in VLDLR/OVR gene weredetected and their association with content of cholesterol in egg yolk was analyzed inthe Silky fowl.First, the egg quality traits were determined and the content of cholesterol in eggyolk were determined by the HPLC. Then, a DNA pool was built using DNA samplesfrom more than100Silky fowls, and sequenced to screen the putative SNPs occurredin the5′-flanking region of the VLDLR/OVR gene in chicken. Then, the PCR-RFLPwas employed to genotype the total312Silky fowls and4SNP loci(g.8355T>C,g.9511G>A,g.9815C>G,g.10428T>CT98C) was found. The associationsbetween the VLDLR/OVR gene and content of cholesterol in egg yolk wasapproaching,which were in order to providing molecular genetic markers forbreeding of low content of cholesterol laying chicken strains.The results are asfollows:1.A total21signal nucleotide polymorphisms were pick out in the in the5′-flanking region of the VLDLR/OVR gene in Silky fowls.2.The genotyping for the2variations found reveled that each of these locioccurred in the population studied with the full3genotypes of4sites.3. The test of Hardy-Weinberg equilibrium showed that g.8355T>C andg.9815C>G locus were not in the balance of Hardy-Weinberg, with a polymorphicinformation content(PIC) of0.324and0.367in the population of interest, respectively.Whereas, g.9511G>A and g.10428T>C locus coincided with the Hardy-Weinbergequilibrium, and PIC were0.349and0.311, respectively. In which the siteg.9815C>G had the most highly polymorphic,biggest mutation degree and bestpotentiality. This indicated that the researched chicken groups had a rich geneticdiversity. 4.The subsequent association analysis of these4loci with the egg quality traitsdemonstrated that g.8355T>C and g.9815C>GT98C have little significationdifference in the content of cholesterol in egg yolk. Whereas, g.8355T>C locusperform significant difference in ESW (P<0.05),and g.9815C>G locus performsignificant difference in EW (P<0.05),YW (P<0.05) and ESR (P<0.01).The results ofrelevance analysis on g.9511G>A and g.10428T>C show that the g.9511G>A locusperform significant difference in YC(P<0.05), YW(P<0.05), and the content ofcholesterol in egg yolk(P<0.01).In which the dominances genotypes were G and GGtype. The g.10428T>C locus perform significant difference in the content ofcholesterol in egg yolk(P<0.05),and the dominances genotypes were T and TT type.
