The reproduction of animals are regulated by complicated factors internally and externally, gonadotrophin (GTH) as the critical hormone in hypothalamic-pituitary-gonadal axis is the key regulatory signal during the development of ovary and secretion of reproductive hormone. Follicle stimulating hormone (FSH) and luteinizing hormone (LH) are steroid hormones secreted from antepituitary, which function is mediated by follicle stimulating hormone receptor (FSHR) and luteinizing hormone receptor (LHR) from follicles and plays a crucial role in development, maturity and ovulation of follicles of female mammals. Healthy unpregnancy mature YiMeng Black Goats were killed for ovaries according to estrous cycle (diestrus, proestrus, estrus and metestrus), real-time qRT-PCR and immunohistochemistry was used to analyse the level of mRNA of FSHR and LHR of ovaries of goats quantitatively and locally. Results showed that:1. RIA was used to show that the concentration of LH in peripheral blood was higher than that of FSH during the whole phases. The concentration of FSH was highest in proestrum, which was significantly different with other phases(P<0.05); and lowest in estrum, which was significantly different with other phases(P<0.05), and the concentration of FSH was not significantly different with metestrus and diestrus(P>0.05). The excertion of LH in peripheral blood was stable and did not have great difference among each phases(P>0.05), the concentration of LH was higher during metestrus and diestrus and lower during proestrus and estrus.2. FSHR mRNA were found in the ovaries of YiMeng Black goats during the four estrous cycle phrases, the amount of FSHmRNA was 0.6187±0.29442(diestrus) ,1.2902±0.29024(proestru), 2.5759±0.45880(estrus),0.7908±0.26193(metestrus) respectively. Expression of FSH mRNA was significantly higher in estrus than other three periods(P<0.01), and lowest in diestrus, which had significant difference compared to proestrus and estrus and had no significant difference with metestrus(P>0.05).3. The results of RT-PCR showed that: LHR mRNA were expressed in the ovaries of YiMeng Black goats during the four estrous cycle phrases, the amount of LH mRNA was 2.2840±0.14622(diestrus),0.5089±0.07899(proestrus),0.3403±0.04538(estrus),2.8592±0.20566 (metestrus) respectively. Expression of LH mRNA was significantly higher in metestrus than other three periods (P<0.01), and lowest in estrus, which had significant difference compared to metestrus and diestrus and had no significant difference with proestrus (P>0.05).4. The results of immunohistochemistry showed that: FSHR positive cells distributed in the ovaries of YiMeng Black goats during the four estrous cycle phrases. FSHR positive cells were mainly distributed in granulosa cells, thecal cells and oocyte, stroma cell, and interstitial tissue of blood vessel were also found positive cells, but were not found in luteal cells.5. The results of immunohistochemistry showed that: LHR positive cells distributed in the ovaries of YiMeng Black goats during the four estrous cycle phrases. LHR positive cells were mainly distributed in granulosa cells, thecal cells, oocyte, interstitial tissue of blood vessel, luteal cells and stroma cell.
