The PRLR gene and steroid BF gene were studied as candidate genes for the prolificacy in two local breeds of Anhui (Wannanhua Pigs and Wannanhei Pigs) in the present study by techniques of PCR and restriction endonuclease. Samples used in this research represented for high prolificacy breeds (Wannanhua Pigs—30 in total;Wannanhei Pigs—85 in total) were obtained from Conservation Farm for Wannanhua Pigs in Huangshanduo County and Conservation Farm for Wannanhei Pigs in Ningguo respectively.(1)As for PRLR gene, one pair of primer was designed to detect single nucleotide polymorphisms of exon 10 of PRLR gene in Wannanhua Pigs and Wannanhchangei Pigs by PCR and digestion with restriction endonucleases according to the sequence of swine PRLR gene. Three genotypes (AA, AB and BB) were detected in both Wannanhua Pigs and Wannanhei Pigs. The gene frequency of AA,AB and BB were 0.667,0.233 and 0.100 in Wannanhua Pigs while in Wannanhei Pigs were 0.5647, 0.2824 and 0.1529. According to statistic analysis, the differences of the litter size in Wannanhua Pigs and Wannanhei Pigs between BB and AA genotypes were all significant (P<0.05).(2)As for BF gene, one pair of primer was designed to detect single nucleotide polymorphisms of intron 1 of BF gene in Wannanhua Pigs and Wannanhei Pigs by PCR and digestion with restriction endonucleases according to the sequence of swine BF gene. Two genotypes (CC and DD) were detected in Wannanhua Pigs (gene frequency were 0.5333 and0.4667 respectively) while Three genotypes (CC, DD and CD) were detected in Wannanhei Pigs(gene frequency were 0.3295,0.5529 and 0.1176 respectively). According to statistic analysis, allele C had a positive heterosis in these two breeds and the differences of the litter size in Wannanhua Pigs and Wannanhei Pigs between CC and DD genotypes, CC and CD genotypes were all significant (P<0.05). These results preliminarily indicated that the PRLR gene and BF gene are potential DNA markers for improving litter size in pigs.
