Lycoris aurea Herb is a perennial herbaceous of Amaryl-lidaceae and Lycoris Herb, it not only has some ornamental value, but also has a wide range of pharmacologically action, particular to its extract galanthamine which can treat myasthenia gravis, polio sequelae and the cure for senile dementia. However, because of the low reproducetion coefficient, unplanned excavation and the backwardness of cultivation techniques, Lycoris wild resources is dwindling. In this paper, Lycoris aurea bulbs were applied for the materials, and two methods that plant tissue in vitro, isolation and fermentation endophyte were to explore the feasibility to produce galanthamine. The main research contents and results are as follows:1. The bulb of Lycoris aurea was used for callus induction, culture me-dium MS+2,4-D2mg/L was singled out for the optimal one to induce callus.2. Callus proliferation medium was studied, and the results showed that MS +2,4-D1mg/L+6-BAO.1mg/L was the optimal medium for callus proliferation.3. Two precursor of Tyramine and 3,4-Dihydroxybenzaldehyde for Syn-thetizing galanthamine were added with the ratio of 1:1 into Lycoris aurea callus suspension culture system, in order to study the influence of the concentration of two precursors on Synthetizing galanthamine. The results showed that adding concentration of 0.05mmol/L can make the highest output of galanthamine.4.10 spezies of endophytic bacteria were isolated from Lycoris aurea bulbs, including 5 bacteria and 5 fungi. After the fermentation culture, one fungus was detected to producing galanthamine with the content of 233.0μg/L. Preliminary identification works showed that this fungus belongs Fungi Imperfecti and Penicillium LK. ex Fries.
