Establishment Of Cell Suspension Cultures And Regeneration Of Plant From Wheat "5389"

In this experiment,wheat cultivar"5389"is cultured to study the effect of differenmediums on callus induction and the gain of embryonal callus and establish the celsuspension culture line of rataria wheat cultivar"5389". The plant regeneration from thcell suspension culture line was also studied. The main results are as follows:1,Effect of different plant growth regulators on callus induction were studiedMS+2,4-D3.00mg/L culture medium can induce wheat cultivar"5389"to produce thhighest ratio of callus.2,Effect of different plant growth regulators on callus subculture were studied.Thconcentration and the proportion of plant growth regulators in subculture medium waregulated with the changing of the callus state. For the eugonic callus we can be reducethe concentration of 2,4-D and increased the concentration of KT properly,with thslow-growing callus need to increase the concentration of 2,4-D and reduce thconcentration of KT properly, but the dosage of plant growth regulators can not be tohigh.By doing this,on MS + 2,4-D 1.00mg/L + KT0.10 mg/L culture medium,thembryonal callus of rataria from wheat cultivar"5389"has been obtained.3,The effect of various concentration of several plant growth regulators on the celsuspension culture line was studied,and the better concentrations were confirmed.Thproduct of the cell suspension culture line was not affected by Casein hydrolysates acid,Glutamine,Asparagine and Malt extract ,but the state of the suspension cell depended othem.4,The stable cell suspension culture line of rataria from wheat cultivar"5389"waestablished by choosing the better culture conditions.The growth curves of suspension celand the cell viability were assayed,then the subculture time of suspension cell wadetermined about 7 days.5,Cell suspension was cultured about 3 month,then take it on culture medium(0.0mg/L NAA and 2.00 mg/LBA) for 3~5 generations,then green bud growing upregenerated plant obtained one month later. In a word,The cell suspension culture line of rataria wheat"5389"was gained and it provided a good system for further studying how the elicitor can induce the defense reaction of host plant.
