Lily(Lilium spp.) belongs to Lilium, Liliaceae. It is a very famous ornamental plant and widely used in food and Pharmacy industry as well. But due to the low propagation rate of lily bulblet and its susceptivity to virus invasion, the product quality of lily remains very poor. Rapid propagation technique can produce large number of plantlets in a short time. Polyploid plants by Chromosome doubling induction have such good characteristics like strong plant, thickened scale etc. For these reasons, the studies on rapid propagation and chromosome doubling of Lily are very important and significant both in theoretic aspect and economic aspect. The paper focused on two main parts, one was the culture medium of rapid propagation of'Siberia'and'Snow Queen'lily; The other was chromosome doubling of'Siberia'and'Pollyanna'and'Snow Queen'lily. The main conclusions were as follows:(l)The ingredients and combination of hormone had a significant effect on the induction of lily bulblet and shoot proliferation of lily. The best suitable differentiation medium of scales of'Siberia'lily was MS+6-BA 1.0 mg/L +NAA 0.5 mg/L, and'Snow Queen'lily was MS+6-BA 1.0mg/L +NAA 0.5mg/L. The best suitable differentiation medium of leaves of'Snow Queen'lily was MS+6-BA 1.0mg/L +NAA 0.5mg/L.(2)Scales in different parts had different differentiation ability. The best suitable differentiation medium of scales at the base of'Siberia'lily was MS+6-BA 1.0 mg/L +NAA 0.5 mg/L.(3)It was better for leaves regeneration of'Snow Queen'lily when bottom of the morphology was downward. The best suitable regeneration medium of leaves was MS+6-BA 1.0mg/L +NAA 0.5mg/L.(4)When the leaves of'Snow Queen'lily used as materials, the best concentration of colchicine was 0.05% with 2%DMSO in chromosome doubling induction of lily, under the concentration, the optimize time was 72h. Compared with the clustered seedlings, leaves were not suitable for polyploid induction.(5)The best time and concentration were different in three kinds of lily.To'Siberia'lily, the best concentration of colchicine was 0.02% with 2%DMSO in chromosome doubling induction of lily, under the concentration, the optimize time was 72h.'Pollyanna'lily was 0.1% with 2%DMSO and the time was 48h.'Snow Queen'lily was 0.02% with 2%DMSO and the time was 48h. So the treatment time and concentration should depend on different varieties on polyploid induction of lily.
