| Fengjie navel oranges(Citrus sinensis Osbeck) is welcomed by consumers for its good tasty and rich nutrition, which contains variety of nutrients of human necessary. In chongqing fengjie, navel oranges in planting area is very wide and is the main economic crops.This experiment was focused on fengjie navel oranges, and the samples were collected from fengjie xian. The main research contents and results as follows:We acquired 14 batches of samples and measured the index of single fruit weight(g),fruit type index,edible rate(%),give juice rate(%),acid amount,reducing sugar,changed sugar,sucrose and total sugar,Vc and sugar acid ratio etc.The extraction technology of flavonoid characteristic ingredients in skin of the fruit of fengjie navel oranges, pulp and juice was optimized, and the best extraction technology was determined. The results showed that methanol was the best extraction solvent and the ultrasonic extraction was the test extraction method. In addition, the content of hesperidin and naringin in defferent parts of the navel oranges was determined. The results showed that the contents of peel higher than flesh and juice.High Performance liquid chromatography method was carried out to study the fingerprint spectrum and its relativity. Results of HPLC indicated that the repeatability of this extraction method was good. The method of fingerprint analysis of Fenjie navel oranges(Citrus sinensis Osbeck) by HPLC has been estabished. Using the reference of hesperidin, the HPLC conditions were optimized as followed:on a DIKMA Diamonsil C18 column, acetonitrile and 0.2% acetic acid solvent was used as the mobile phase, the UV detection wavelength was 290 nm, the column temperature was 30℃, and the velocity of floe was being 1.0ml/min.14 batches of Fengjie navel oranges(Citrus sinensis Osbeck)were analyzed with the HPLC fingerprint method. With the standard substance of hesperidin, the similarity of all above chromatograms were compared and the fingerprints of Fengjie navel oranges were built. The spectrum gotten in this experiment showed that the chemical fingerprint information is overall, and the resolution of the peaks is good with obvious characteristics.The precision, stability and repeatability of this method meeted the requirements of the research on fingerprint spectrum. |
