Herbaceous peony (Paeonia lactiflora Pall) is China’s traditional flowers which has high ornamental values. Two aspects of embryo and stem segment callus culture were studied in this experiment, mainly studies effect of different embryonic age on young embryo culture, the embryo initiation, axillary shoot induction and proliferation, rooting, acclimatization and transplant, callus induction, proliferation and differentiation of five varieties which was’Da Fu Gui’,’Sorbet’,’Coral Sunset’,’Duchess de Nemours’and’Sarah Bernhard’. Main results were as following:1. The effect of initiation was best of mature embryo (after flower90d). Embryo removed seed coat and endosperm was the best explants treatment.2. The best combination of hormone for embryo initiation was6-BA0.5mg/L+GA31.0mg/L.3. The best way for axillary shoot induction and proliferation of seedling induced by embryo was:first, seedlings were transferred to1/2MS (double-strength Ca2+)+6-BA1.0mg/L+GA30.5mg/L for cultivation for20d, the optimum medium for axillary shoot proliferation was1/2MS (double-strength Ca2+)+6-BA1.0mg/L+GA31.0mg/L.1/2MS without PGRs was the best medium for strong root.4.1/2MS was the best basic medium of callus induction for’Coral Sunset’,’Duchess de Nemours’and’Sarah Bernhard’; the optimum induction medium was TDZ0.5mg/L+NAA0.5mg/L for’Sorbet’and’Sarah Bernhard’, the best induction medium of’Coral Sunset’and’Duchess de Nemours’was TDZ0.5mg/L+2,4-D0.5mg/L.5. The most suitable medium for callus proliferation of’Du Fu Gui’was2,4-D0.5mg/L+TDZ0.5mg/L or2,4-D1.0mg/L+TDZ0.5mg/L+NAA0.5mg/L;2,4-D1.0mg/L+TDZ0.5mg/L was the best callus proferation medium for’Sorbet’; TDZ0.2mg/L+NAA0.2mg/L was the appropriate proferation medium for’Coral Sunset’; the best callus proferation medium for ’Duchess de Nemours’ and ’Sarah Bernhard’ were2,4-D1.0mg/L+TDZ0.5mg/L+NAA0.5mg/L and2,4-D1.0mg/L+TDZ0.5mg/L+NAA0.2mg/L respectively.6. The optimum differentiation medium for’Da Fu Gui’and’Sorbet’were WPM+NAA0.5mg/L+TDZ0.5mg/L and WPM+NAA0.5mg/L+TDZ0.2mg/L respectively; darkness culture conductived to differentiation of’Da Fu Gui’ and could inhibit browning and pollution effectively; the appropriate CH could promote callus differentiation. The best differentiation medium were1/2MS+TDZ0.5mg/L+NAA0.5mg/L,1/2MS+TDZ0.5mg/L+NAA0.5mg/L and1/2MS+TDZO.1mg/L+NAA0.2mg/L for’Coral Sunset’,’Duchess de Nemours’and ’Sarah Bernhard’ respectively. The liquid culture way was more conductive to callus differentiation of’Sarah Bernhard’.7.When IBA was1.Omg/L, callus grow very well of’Coral Sunset’、’Duchess de Nemours’ and’Sarah Bernhard’, but did not induce the formation of adventitious roots in the end.8. Prolonged culture period and1/2MS could be partly restored browning callus.
