Cordyceps militaris is a medical fungl of famous tradition, with abundant nutrition and prenominent medical function. Now, the research about Cordyceps militaris is most foucs on cordycepin and polysaccharides. So the solid culture, fermentation medium and condition, extraction of carotenoids and extraction of flavonoids were studied on the Cordyceps militaris.(1) The best Cordyceps militaris was separated from escape Cordyceps militaris and selected by solid growth experiment. The sorophore of SY12 was best in yield and mass, average growth cycle 45d, average length 9.10cm, average diameter 0.25cm, fresh weight 31.71g, and excellent heredity and stability from continual experiments.(2) The fermentation conditions of Cordyceps militaris were studied by single factor and orthogonal experiments. The results of experiments showed that the optimal fermentation medium was composed of sucrose 4.0%, peptone 3.0%, (NH4)2SO4 0.6%, K2HPO4 0.05%, and MgSO4 0.02%. The optimum fermentation conditions were fermentation temperature 25℃, initial pH7.0, inoculums volume 8%, fermentation time 96h. The maximum biomass of 18.7g/L was obtained under the optimized conditions.(3) The experiment results showed that the method combining hydrochloric acid (HCl) with heating (by boiling water) was suitable for extraction on the carotenoid produced from Cordyceps militaris. Acetone was the optimal extracting organic solvent. The optimum extraction process was selected with the orthogonal design. The optimal extraction condition was obtained as follows: HCl concentration 1mol/L, HCl addition 15mL/g (dry sample), soaking time 20min and heating time 4min. Under the above optimal conditions, the yield of carotenoid could reach 2158μg/g dry sample. The peak values of UV-VWAS spectrum were 415nm, 439nm, 468nm respectively, according to the eigenvalue carotenoids.(4) The results of experiment show that the ethanol refluxing was optimum for extraction on the flavones produced from Cordyceps militaris. The optimum extraction process was selected with the orthogonal design. The optimal extraction condition was obtained as follows: ethanol concentration 75%, ratio of liquor and material 40:1, refluxing time 60min and extraction twice. The quantity of flavones was also basically identified by color reaction and UV-VWAS spectrum. The flavonoids could be determined preliminary from Cordyceps militaris.The study on extraction of carotenoids and flavonoids from Cordyceps militaris provided important reference for the qualities analysis and medical value.
