This study focused on Cordyceps militaris, a famous traditional Chinese edible and medicinal mushroom, It has a very high utilization value. We collected 4 wild Cordyceps militaris from Fushun. The optional culture requirements for Cordyceps militaris fruit body and mycelium were investigated, in order to established an integrated method of good strains selection, and using the strain selected as the parent strains to carry out anti-aging effects of Cordyceps militaris.First of all, the different strains were cultured in different nutritional conditions (carbon source, nitrogen source, proportion of nutrient liquid and material, etc) and environmental factors (temperature, humidity, light, venting, etc) that were isolated from wild Cordyceps militaris, and the different training methods (rice solid medium, subsurface fermentation, etc) were used for Cordyceps militaris cultivation, and then we analyzed functional components (cordycepic polysaccharide, amino acid, cordycepin, cordycepic acid, SOD, etc) of different Cordyceps militaris samples. We established an integrated strains selection method and an optimal cultivation method of Cordyceps militaris.The results indicated that No.1 was the good strains with high yield of fruit body and mycelium and high content of functional components. It is the optimal culture conditions for fruit body of Cordyceps militaris:the optimal carbon source was starch and the optimal nitrogen source was peptone, the optimal proportion of nutrient liquid and rice was 1:1, the optimal culture temperature was at 22℃-25℃and the optimal relative humidity was at 65%-70%, the mycelium of Cordyceps militaris could well grow in the dark. Given illumination stimulation after the mycelium had had enough culture medium, which would guide the fruit body's forming. The optimal culture temperature was at 13℃-25℃and the optimal relative humidity was at 70%-75%, venting, lighting and temperature stimulation would promote the growth of fruit body. It is the optimal culture conditions of subsurface fermentation for mycelium of Cordyceps militaris:the optimal carbon was glucose and the optimal nitrogen source was peptone, the inoculation amount of liquid trains was 5%, the cell age of liquid trains was 48h, the filling liquid of fermentation bottle was 100ml/250ml, initial pH was 7.0-7.5, the shaker rate was 150 r/min, the culture temperature was 22℃-24℃, the fermentation time was 76h.Selected the No.1 strain as the parent strains to culture fruit body and mycelium, and used the fruit body and mycelium to study anti-aging effects of Cordyceps militaris. The endurance test, the reaction sensitivity test and the germ cell viability test were carried out with mice, the oxygen free radicals removal test was carried out with old rats, and studied the scavenging action of hydroxyl free radical of the wild and the cultivated Cordyceps militaris. The results indicated that the cultivated Cordyceps militaris has obvious anti-aging effects and good prospects for development.
