IGF-I, insulin-like growth factor can regulate organism growth, development and metabolism, and is mediator of GH realizing bioactivity. IGF-I is a single copy gene, while due to alternative splicing, the use of multiple polyadenylation signals and different promoters, the IGF-I gene and its mRNAs are complicated therefore. As a multiple candidate gene, IGF-I gene is the key regulates animal reproductive gene, which mainly relies on GH/IGF-I growth axis to react to reproductive system. This research applies to reproductive organ of Junmu 1 boar and broad sow in different developmental stages as the studying object, analyzes the expression features in pig reproductive organ by different splicing type of IGF-I total mRNA and IGF-I, which lays theoretical foundation of thorough study of reproductive theory regulation.This experiment analyzes the expression features of IGF-I total mRNA on testicle, uterus and ovary in different developmental stage, and gets following results: 1. IGF-I total mRNA can express its feature in each developmental stage of Junmu 1 testicle, especially in sexually matured developmental stage, and IGF-I total mRNA regulates the sexually matured development of boar.2. IGF-I total mRNA can express its feature in each developmental stage of Junmu 1 uterus and ovary, especially in estrus cycle of sexually matured broad sow, and IGF-I total mRNA regulates the estrus cycle of boar. IGF-I is one of key alternative genes in developmental and reproductive progress of Junmu 1 boar and broad sow, especially in the sexually maturation period and after-sexually maturation period.Because of different alternative splicing, polyadenylation signals and potentially multiple promoters, the IGF-I gene builds different type of mature mRNA splicing structure by extremely complicated way. This research takes Junmu 1 pig as the research object, applies semiquantitative RT-PCR method, and finally achieves expression features of four transcriptons sequences, the I-Eb type, I-Ea type IGF-I and II-Eb type, II-Ea type, so as to identify the mainly splicing type of four transciptons, which one has the greatest effect to animal reproductive organs, in different developmental stage of testicle, uterus and ovary. The results show, I-Ea type IGF-I keeps high expression. The expression rule of four IGF-I splicing types in different developmental stage of uterus:The uterus expression features of IGF-I I-Eb type are obviously higher than the others,. The expression rule of four IGF-I splicing types in different developmental stage of ovary:Four IGF-I splicing types have each expression rules. It is hard to define the key effective IGF-I splicing type. Thus, we need further study.
