Objective:In this study, to investigate the effect of Forsythoside A inhibiting avian infectious bronchitis virus (IBV) in vitro,the chick embryo kidney (CEK) cell culture model was established, and The expression of IFN-αand related factors of JAK-STAT signaling pathway were investigated when forsythoside A put on CEK cells.Methods:The standard strain of IBV-M41 was used to infect chicken embryo kidney cells, to detect the effect of forsythoside A inhibit IBV the content of IBV N gene was detected by real time quantitative PCR and cytopathic (CPE) observation; And, to judgement the effect of forsythoside A inducing the expression of IFN-a and factors of JAK-STAT signaling pathway, the expression of IFN-a and related factors of JAK-STAT signaling pathway were observed by real-time quantitative PCR, furtherly, the protein expression levels IFN-a was determined by western-blot in this study.Results:The results show that when IBV treated with forsythoside A in the concentration of 0.64mM directly,it can inhibit IBV completely after 36h, and 0.64mM forsythoside A pretreatment CEK cells can inhibit IBV partially; and compared with the normal group, produced the expression of IFN-a, and related factors of JAK-STAT signaling pathway were detected after high-dose forsythoside A was added to CEK cells at 12 hours.Conclusion:When forsythoside A in a certain concentration range, the replication of IBV can be inhibited, and it can also induce IFN-a and regulate the JAK-STAT signaling pathway positively.
