Effects of Bacillus sp.andβ-Mannan as feed additive on intestinal incretion and immunity of rabbit was studied in this study.Forty-eight California Rabbits(male) were divided into 4 groups in this study after equipped breed:Group 1 with basic diet,Group 2 with basic diet and 0.1%PAS38,Group 3 with basic diet and 0.15%Mannan,Group 4 with basic diet and 0.1%PAS38+0.15%Mannan.4 Rabbits in each group were killed after weigh per time-point on day 15,30,45-day-old.Set out their operative capability,the distribution and change of IEL,somatostatin(SS)-positive cells,IgA-positive cells,5-HT-positive cells and mast cells in duodenum and jejunum of rabbits were studied.1.Metage avoirdupois of rabbits per time-point on day 15,30,45-day-old.Reclaim and metage feedstuff everyday.Count dayly average weight gain,appetite and appetite/weight.The result showed that Bacillus sp.andβ-Mannan could improve dayly average weight gain and appetite/weight of rabbits.The effect was better when they were both used combine.2.H.E staining method was used to study the change of mucosa thickness and villus length in rabbit small intestinal.The results showed that Bacillus sp.andβ-Mannan as food of rabbits to increase mucosa thickness and villus length in rabbit small intestinal inceased.The effect ofβ-Mannan was better than Bacillus sp.on prophase beed.But the effect of Bacillus sp was better on anaphase feed.The cooperate domino effect was showed when they were used combine.3.H.E staining method was used to study the change of IEL in rabbit small intestinal.The results sho-wed that Bacillus sp.andβ-Mannan as food of rabbits to increase the number of IEL in rabbit small intestinal increased,the number of IEL in duodenum of mix group increased significantly(P<0.05) compared to control group at 15day;At 30 day,comparing to control group the number of IEL in duodenum of Bacillus sp.group andβ-Mannan group increased significantly(P<0.05),mix group increased significantly(P<0.01);the number of IEL in jejunum of Bacillus sp.group and mix group increased significantly(P<0.05) compared to control group at 30 day;At 45 day, the number of IEL in duodenum of Bacillus sp.group and mix group increased significantly(P<0.05) compared to control group.Our results suggest Bacillus sp.andβ-Mannan as food of rabbit increase the number of IEL in rabbit small intestinal increased.And the effect of mix group was best. 4.The changes of the number of IgA-positive cells in rabbit small intestinal were studied by SABC immunohistochemistry.At 15 day,the number of IgA-positive cells in duodenum of mix group increased significantly(P<0.05) compared to control group;At 30 day,the number of IEL in duodenum of Bacillus sp.group andβ-Mannan group increased significantly(P<0.05) compared to control group,and mix group increased significantly(P<0.01);the number of IgA-positive cells in jejunum of all experiment group increased significantly (P<0.05) compared to control group at 30 day;At 45 day,the number of IgA-positive cells in duodenum of mix group increased significantly(P<0.05) compared to control group.Our results suggest Bacillus sp.andβ-Mannan as food of rabbits to increase the number of IgA-positive in rabbit small intestinal increased,thus mucosal immunization of small intestinal was promoted.The effect of mix group was best.5.The changes of the number of 5-HT-positive cells in rabbit small intestinal were studied by SABC immunohistochemistry.At 15 day,the number of 5-HT-positive cells in duodenum of Bacillus sp.group and fl-Mannan group increased significantly(P<0.05) compared to control group,the mix group increased significantly(P<0.01);the number of 5-HT-positive cells in jejunum of Bacillus sp.group andβ-Mannan group increased significantly(P<0.05) at 15 day compared to control group,and mix group increased significantly (P<0.01);At 30 day,the number of 5-HT-positive cells in duodenum of Bacillus sp.group and mix group increased significantly(P<0.01) to control group;Comparing with control group,the number of 5-HT-positive cells in jejunum of Bacillus sp.group andβ-Mannan group increased significantly(P<0.05) at 30 day,the mix group increased significantly(P<0.01);At 45 day,the number of 5-HT-positive cells in duodenum of mix group increased significantly(P<0.05).Our results suggest Bacillus sp.andβ-Mannan as food of rabbits to increase the number of 5-HT-positive cells in rabbit small intestinal increased,thus the function of incretion and immunity in rabbit small intestinal was boosted up.6.Toluidine blue staining method was used to study the change of mast cells numble in rabbit small intestinal.The results showed that the number of mast cells in duodenum of mix group increased significantly (P<0.05) at 15 day compared to control group;the number of mast cells in jejunum of Bacillus sp.group and mix group increased significantly(P<0.05) compared to control group at 15 day;At 30 day,the number of mast cells in duodenum of Bacillus sp.group andβ-Mannan group increased significantly(P<0.05) compared to control group, mix group increased significantly(P<0.01);the number of mast cells in jejunum ofβ-Mannan group increased significantly(P<0.05) compared to control group at 30 day,Bacillus sp.group and mix group increased significantly (P<0.01);At 45 day,the number of mast cells in duodenum of mix group increased significantly(P<0.05) compared to control group;the number of mast cells in jejunum of Bacillus sp.group increased significantly (P<0.05) compared to control group,mix group increased significantly(P<0.01).Our results suggest the number of mast cells might reflect the digestive mucosal immunization.7.The changes of the number of SS-positive cells in rabbit small intestinal were studied by SABC immunohistochemistry.At 15 day,the number of SS-positive cells in duodenum of Bacillus sp.group reduced significantly(P<0.05) compared to control group;the number of SS-positive cells in jejunum of all experiment group reduced significantly(P<0.05) compared to control group at 15 day;At 30 day,the number of SS-positive cells in duodenum of Bacillus sp.andβ-Mannan group reduced significantly(P<0.05) compared to control group,mix group reduced significantly(P<0.01);the number of SS-positive cells in jejunum ofβ-Mannan group reduced significantly(P<0.05) compared to control group,Bacillus sp.group and mix group reduced significantly(P<0.01);At 45 day,the number of SS-positive cells in jejunum of mix group reduced significantly (P<0.05) to control group.Our results suggest Bacillus sp.PAS38 andβ-Mannan as food of rabbits to boost up the growth capability, incretion and immunity of small intestinal.The effect was best when they were both used combin.
