| With the intensification of aquaculture as well as condition deterioration in China,there have been significant increases of various kinds of aquatic diseases.To prevent or treat fish diseases,a substantial quantity of antibiotics and chemicals are used,which caused a lot of problem such as drug residue and drug resistance,therefore the search for the effective and safe green drugs for aquaculture has become the focus of increasing attention.The objective of the research was to probe the hepatoprotective aspect of baicalin against the carbon tetrachloride(CCl4)-induced liver injury in crucian carp using the technique of precision-cut liver slices(PLCS),so as to provide therietcal bases for the use of baicalin as hepatoprotective agent.8mM CCl4 was used to establish the injury model of PLCS in crucian carp.Different control and treatment group were set as follow:blank control group(without any drugs),model group(CCl4),baicalin control group,baicalin(0.05,0.1 and 0.2mg/ml)treatment groups(pre-treatment group,post-treatment group,and pre-and post-treatment group).The supernatant and homogenate of liver slices in each group were collected and prepared to detecte total antioxidant capacity(T-AOC),alanine transaminase(GPT),catalase(CAT),aspartate transaminase(GOT)alkaline phosphatase(AKP),superoxide dismutase(SOD),lactate dehydrogenase(LDH),total protein(TP),albumin(Alb),malondialdehyde(MDA),peroxidase(POD),glutathione transferase(GST),glutathione peroxidase(GSH-Px),adenosine triphosphate(ATP),cytochrome P450(CYPs)(CYP1A,CYP3A),interleukin-1β(IL-1β),nuclear factor-KB(NF-κB),interleukin-6(IL-6),inducible nitric oxide synthase(iNOS).Studies displayed that contrasted with the blank,baicalin at the medium-dose and high-dose in pretreatment group and pre-and post-treatment group displayed the better effect on the protection of liver slices,and the other experimental groups also showed effect on the inhibition of the CCl4-induced liver injury.Baicalin significantly reduced the levels of GOT,GPT,AKP,LDH and MDA,increased the activities of antioxidant enzymes such as SOD,T-AOC,CAT,GSH-Px,GST,and increased the contents of TP,Alb and ATP in liver slices.Moreover,baicalin significantly inhibited the gene expressions of CYP1A,CYP3A,c-Rel,IL-1β,IL-6,iNOS.DPPH assay showed that baicalin even at low concentrations had potent radical scavenging activity.Conclusion:Baicalin can effectively reduce the fish liver damage by CCl4,and inhibited lipid peroxidation and inflammatory response.Baicalin may potentially be used as a hepatoprotective agent for fish liver disease. |
