Muskmelons (Cucumis melo L.) demonstrates remarkable hybrid vigor, almost all melon seeds are hybrids cultivars in melons production. Using monoecious lines as F1 hybrid parents is a potentially stable and effective means to reduce high cost of hand pollination and solve the problem of stamen packing by labor. It could also improve purity of hybrid seeds. Through selective breeding , two melon monoecious lines ATA-A and ATA-B were received by Watermelon and Melon research group of Horticulture Department of Northwest A&F University. Using monoecious lines ATA-A, ATA-B and an andromonoecious line and their offsprings (F1 and F2 and BC1) as materials, heredity regular and molecular marker of monoecious character were studied. The results are following:1. Flower sex expression of monoecious lines (ATA-A and ATA-B) and andromonoecious lines and their offsprings (F1 and F2 and BC1) was investigated. Results showed that the flower expression character of that monoecious lines of melon was controlled by a pair of genes at one single locus,and the Monoecious character was controlled by the dominant gene.2. In the study on the optimization of RAPD reaction system for melon, using genomic DNA of melon as the template, the concentrations of Mg2+, dNTPs, DNA, random primers and Taq DNA polymerase were tested. The results indicated that the optimized reaction system was :25μL as total, 2.0mM的Mg2+,0.2mmol/L的dNTPs,primer 0.2μmol/L,30ng DNA,Taq DNA polymerase 1.0 U. This system had the traits of clear, good polymorphic and well repeated electrophoresis patterns. A stable PCR program was obtained through single factor test.3. 80 individuals of ATA-A F2 plants were used for RAPD molecular mapping of the monoecious gene by bulk segregant analysis (BSA). 840 random primers were chosen for polymerase chain reaction (PCR) amplification between monoecious pool and andromonoecious pool, the results showed that: primer S128 was tightly linked to the monoecious character, and primer S493 was tightly linked to the andromonoecious character.4.
