Nangyang cattle (aged 2 years old) and Qin Chuan cattle (aged 4 months),totally 163 beef cattle, were used as research populations. The DNA molecular marker technologies, such as PCR-RFLP, PCR-SSCP, were applied to evaluate the effects of genotypes of three candidate genes on growth traits in Nanyang cattle and also to detect genetic diversity of POU1F1, Myf-5 and GH genes in the populations. The Association between genetic marker and the performance traits, including the body weight, body length, heart girth, withers height, height at hip cross, hip width, rump length and ischium width were analyzed using general linear model (GLM). The results were as follows:1 Association of polymorphisms of POU1F1 gene with growth traits in Nangyang cattlePCR-RFLP was firstly applied to analyze the association of variations of POU1F1 gene with growth traits in 100 Nanyang cattle. The results showed that 451 bp PCR products digested with HinfI demonstrated polymorphisms in population, which was at Hardy-Weinberg equilibrium. Moreover, the frequencies of alleles A/B in Nanyang population were 0.465/0.535. Association of variations of POU1F1 gene with growth traits was analyzed. In birth weight, average weight increase before ablactation, body height at 12 months, body weight, body length and chest girth at 6 months and 12 months, individuals with genotype BB were higher than that with genotype AB in Nanyang population, that was BB>AB(P<0.05); In body weight at 12 months, individuals with genotype BB were higher than that with genotype AA in Nanyang population, that was BB>AA(P<0.05), although there was no significant difference in other traits among individuals with the two different genotypes in Nanyang population, the trend of allele B>allele A.2 Association of variations of MYF-5 gene with growth traits in Qinchuan cattle and Nanyang cattlePCR-RFLP was firstly applied to analyze the association of variations of MYF-5 gene with growth traits in 163 cattle, which comprised of Qinchuan cattle, aged 4 months(±10 d)in the same season, and Nanyang cattle, aged 2 years.
