POU1F1 (pituitary transcription factor 1)gene acts as a positive regulatory factor of GH,PRL and TSH-β, and plays an important role in the initiation of expression of related genes and regulation of early growth in mammal. Therefore, it is selected as a candidate gene for growth trait. In this study, a 2.6kb POU1F1 5'flank region was cloned firstly,then the distributaion of polymorphisms in 8 pig breeds were evaluated, and at last we studied the association between different genotypes of POU1F1 5'flank region and growth performance in Sujiang pig .1 Cloning and sequence analysis of the 5'Flanking region of Porcine POU1F1 gene.A 2.6kb POU1F1 5'flank region was cloned firstly by Gemone Walking approach from Landrace genomic template and sequenced then. The result had been submitted to Genebank (No. EU924264).Then the sequence was analyzed with biology software and alignmented with the POU1F1 5'flanks of others animal to construct the phylogenetic tree.The result revealed that the cloned region contained typical TATA box(-57) and CAAT box like sequence(-87). The alignment results showed that the conservative region spaned from -150 to -1000bp of upstream of transcription start site,and the -150 bp upstream of transcription start site was strongly conservative and it was potential core promoter. The phylogenetic tree constructed by MLAGAN program of Vista represneted the evolution relationship among different species.2 Polymorphisms detection in the 5'Flank of pig POU1F1 gene in 8 porcine breedsFive polymorphisms had been identified by comparative sequencing the Yorkshare and Xiang pig POU1F1 5'flank region.It included four mutation site and one deletion/insertion site (ggagtg). The deletion/insertion mutation contains a poteintial transcription factor (GAGA and TFII) binding site. The distributaion of deletion/insertion (ggagtg) polymorphisms in 8 pig breeds(Lnadrace, Yorkshire, Duroc, Sujiang, Tibet pig, Rongchan pig, Erhualian and Meishan pig) were evaluated by PCR-RFLP-Hpy188 I. Chi-square test showed that the genotypic ferencies of every pig breed were accordrace with the Hardy-Weiberg balance. Different distribution of genotypic ferequencies were observed in different breeds. In Yorkshire, Duroc, Landrace, the genotype AA was higher, the genotype AB was much less, and no genotype BB was detected in them. All of three genotype AA,AB,BB appeared in Meishan pig, Erhualian pig,Sujiang pig and Tibetan pig appeared .3 The relationship between polymorphism of porcine POU1F1 gene and the growth performance in Sujiang pigs.The result of the association between different genotypes of POU1F1 5'flank region and growth performance in Sujiang pig showed that individuals with genotype AA were significant higher than that with genotype AB and BB in body length ,body weight and chest depth (P<0.05) ,although there was no significant difference in backfat thickness among individuals with the three different genotypes (P>0.05). These results indicate that the allele A has a favorable positive effect on growth traits.
