PCR-RFLP and PCR-SSCP methods were used to analyze the HinfI,AluI,PstI loci in the Exon6 of POU1F1 Gene and the Exon1,Exon2,Exon3,Exon4 of LPL gene in 311 Jiaxian Red cattle,Qinchuan cattle and Nanyang cattle.The Association between the polymorphisms and the body weight and body size were analyzed,which aimed to explain its molecular genetic information,find effective molecular markers associated to economic traits,and gave scientific theoretical basis to the protection and utilization of Chinese cattle,the developing of the molecular marker assistant selection(MAS) breeding and the improvement of the cattle performance.The results were displayed as follows:1.The association between the polymorphisms of the Exon6 of POU1F1 gene and the growth and development traits in Jiaxian Red cattle451 bp PCR products digested with HinfⅠdemonstrated polymorphisms in analyzed population which were at Hardy-Weinberg disequilibrium(P<0.05).Moreover,the frequencies of allele AA,AB and BB genotype of Jiaxian Red cattle were 0.1875,0.3056 and 0.5069,accordingly,the frequencies of allele A and B were 0.3403 and 0.6597,respectively.The homozygositiy,heterozytosiy,the effective allele numbers,and PIC value of POU1F1-HinfI locus were 0.5510,0.4490,1.8148 and 0.3482,respectively.As this locus belonged to the middle polymorphic index,it was suggested to become a effective evaluation parameter for studying genetic resource.However,no polymorphism was found at POU1F1-AluI and POU1F1-PstI loci.The correlation of HinfI polymorphisms of POU1F1 gene and growth traits was analyzed and the result showed that there was significant relationship between HinfI polymorphism and body lengthen(P<0.05).The individual with genotype BB has longer body length than the individual with genotype AB,suggesting the allele B had significant effect on body length of Jianxian Red cattle.The genotype BB significant correlation with the fine performance and the selection to the allele B is better for the improvement.2.The association between the polymorphisms of the LPL gene and the growth and development traits in Qinchuan cattleThere were polymorphisms in the Exon2 and Exon5 of the LPL gene in Qinchuan cattle analyzed by PCR-SSCP,which were contrary to the Exon1 and Exon3.(1)Three genotypes(AA,AB and BB) were detected in the LPL-Exon2 locus in Qinchuan cattle,which had no influence to the body size.The allele frequencies of A and B were 0.6855 and 0.3145,respectively,which were under Hardy-Weinberg equilibrium(P>0.05).(2)The gene frequencies of A and B in LPL-Exon5 were 0.3226 and 0.6774, respectively,the PIC was 0.3415 belonged to middle polymorphism,and population were under Hardy-Weinberg equilibrium,too.The individuals with AB genotype had better body height,height at hip cross,body length,heart girth,rump length and body weight than the individuals with AA and BB genotype significantly(P<0.05).3.The association between the polymorphisms of the LPL gene and the growth and development traits in Nanyang cattleThere were polymorphisms in the Exon2 and Exon5 contrary to Exon1 and Exon3 of LPL gene analyzed by PCR-SSCP in Nanyang cattle.(1)The genotypes of A and B in the LPL-Exon2 were 0.7095 and 0.2905 respectively,and the heterozygosity and PIC were 0.4122 and 0.3272 respectively.The population was under Hardy-Weinberg equilibrium.The association between the genotype and the growth and development traits showed that:The genotype AB had significant influence to the birth weight and the body height in 18 months(P<0.05).(2)There were three genotypes(AA,AB and BB) in the LPL-Exon5 locus,the heterozygosity and the PIC were 0.4638 and 0.3562,respectively,and the population was under Hardy-Weinberg equilibrium.The individuals with AB genotype had better body weight and body size than the individuals with AA genotype,but the difference wasn't significant,while the individuals with AB genotype had better birth weight than the homozygotes(P<0.05).
