Pituitary transcription factor 1 (POU1F1) can recognize and combine with the DNA target sites to control transcription and gene expression of target Genes such as growth hormone (GH) gene, Prolactin (PRL) Gene and thyroid hormonesβ(TSHβ) gene, have a vital role to play in the growth, development, reproduction and immunization of the animals. In this study,100 Tianfu goats,40 Bore goat and 38 Chengdu Ma goats were used to analyse the farm effect between the two farms of the Tianfu goat stud and identify POU1F1 gene polymorphism by PCR-SSCP, and analyse the association of the different genotypes with the POU1F1 genetic effects in Tianfu goats. Hope to find out some polymorphism sites, which closely relate to growth and production traits, to provide scientific basis for the breeding practice of Tianfu goat by marker assisted selection (MAS). The results were in the followings:There were two mutations in the intron 4 region of POU1F1 gene in Tianfu goats and Bore goat, IVS4+528A>G and IVS4+708delA. They produced three types AA, AB and BB by PCR-SSCP, and the genotype frequencies were 0.76,0.15,0.09 and 0.325,0.40,0.275, respectively. A allele was an advantage gene. There was no polymorphism in Chengdu Ma goats.The average polymorphic information contents (PIC) were separately 0.238 and 0.411. Chi-square analysis indicated that the loci of POU1F1 detected deviated from the Hardy-Weinberg equilibrium in Tianfu goat and Bore goat(P<0.05).Analysis of the farm effect showed the two farms of the Tianfu goat stud had no significant difference between growth traits and reproduction traits (P>0.05).According to results of the POU1F1 genotypes effect analysis for growth traits showed that the birth weight and 2 months body weight of AA genotype were much significantly higher than BB (P<0.01), and obviously higher than AB (P<0.05), There were no significant difference between AB and BB (P>0.05); 2 months body length, body hight, chest girth, and 4 months body weight, body length, body hight,and 9 months chest girth of AA genotype were significant higher than BB (P<0.05), and no significant difference between AA and AB (P>0.05). And there were no significant difference between AB and BB (P>0.05) except that 4 months body hight of AB was greater than BB (P<0.05). In the others growth stages, there were no significant difference with the others growth traits between the three of genotypes (P>0.05). It suggest the polymorphic can be the candidacy molecular marker for the early growth of the Tianfu goat. The reproduction traits of different POU1F1 genotypes had significant difference in the first litter and the total number of the first three litters in Tianfu goat, they were all BB>AA (P<0.05), and comparison between AB and AA or BB had no significant difference (P>0.05). |
