Cordyceps ophioglossoides, a rare and valued traditional Chinese medicine, belongs to Cordyceps, Clavicipticeae, Hypocreales, Ascmycotina. According to the recordation of " Xinhuabencaogaiyao" , this medicinal fungus is used to cure Menoxenia and can enhance the immunity. However, Cordyceps ophioglossoides as a natural resource is now closing to die out due to environment deterioration, so artificial cultivation is an effective method to retrieve this organism. In this work, the liquid culture conditions of C. Ophioglossoides for the production of mycelial biomass were investigated in 15L agitatition fermentor, and the active, components of C. ophioglossoides mycelia were analyzed.The strain C ophioglossoides Li01 which was isolated from the wild fruiting body of the fungus in Yunnan. The basic culture medium was 66g/l of sucrose, 10g/l of yeast extract, 3.0%(v/v) of silk worm hydrolysate, 0.4g/l of MgSO4·7H2O and 0.4g/l of KH2PO4. For optimizing fermentation condition, the orthogonal layout L9(43) was carried out in 15 L agitatition fermentor under the conditions of the primary aeration rate as 0.6vvm and agitation rate as 150rpm, in which the four main influence factors were selected as test factors including inoculum volum, temperature, initial pH, metaphase aeration rate. The experimental results indicated that the optimum fermentation conditions was 5% of inoculum volum temperature 23℃, pH5.5 and aeration rate changed to 1vvm after 40h. Under the optimum fermentation conditions above, the 9.5g/l of maxium mycelial biomass was achieved, which is about 2 times higher than that at the basic fermentation conditions.In addition, the analysing method of the active components were established for C. ophioglossoides mycelia, and the result was revealed to be 14.06mg/g of polysaccharides, 58.89mg/g of mannitol 74.79mg/g of proteins 2.17mg/g of adenosine under the optimal culture conditions.
