Silkworm Cordyceps is a silkworm-fungus complex that the Cordyceps militaris infected Silkworm body and produced the fruiting body. The Silkworm is chosen to be the host due to its easy raising, large breeding scale and the most similar morphology to the worm of wild Cordyceps sinensis. Both Silkworm and Cordyceps militaris have high medicinal value; therefore, the cultivated Silkworm Cordyceps might be the best substitution of wild Cordyceps sinensis. This study chose the silkworm to be the host, screened the best strains for infecting dead silkworm and live silkworm, optimized the treatment and growth condition for dead silkworm, built the processing route for cultivating live silkworm cordyceps in stress and detected the main active constituent of the silkworm cordyceps.1. It is found that the best strain infecting dead and live silkworm is C23and C22, respectively, by screening investigation. At the same time, the best inoculation methods—injecting liquid strain was screened.2. The cultivated route of the dead silkworm cordyceps was built by optimization of the treatment of the dead silkworm and growth condition of the dead silkworm cordyceps. The results showed that the best treatment methods of dead Silkworms is drying at100℃for30min and then sterilizing at high temperature and high pressure. The optimal temperature and humidity for cordyceps hyphae growth is23℃and70%, the best for fruiting body growth is temperature23℃, humidity90%and intensity150lx.3. The live silkworm cordyceps cultivation route was established by optimization of cultivated conditions. The best disinfection methods of living silkworms body surface was ultraviolet disinfection for30min, afterwards, cultured at7℃for8days, injected0.3mL liqid strain, cultured on the soil with15%water. The average infection rate reached89%with two repeat.4. The main active ingredients of three parts (fruiting body, worm body, whole body) of Silkworm Cordyceps were detected and analyzed, and compared with that of Cordyceps sinensis and Cordyceps militaris. Results showed that most of the main active ingredients were located at the fruiting bodies. Cordycepin of fruiting body was about2.45times of Silkworm body and polysaccharide was about2.98times of Silkworm body and cordycepic acid was about5.45times of silkworm body. Comparison analysis between silkworm cordyceps with Cordyceps sinensis, Cordyceps militaris found that cordycepin and polysaccharide of the silkworm codyceps fruiting body is the highest among these three. Cordycepin of fruiting body is about2.98times of fruiting body of cordyceps sinensis and1.73times of Cordyceps militaris fruiting bodiy. The polysaccharide content of Silkworm Cordyceps fruiting body is the highest Its content is about6.61times of the Cordyceps sinensis fruiting body and about1.52times of Cordyceps militaris fruiting bodies. However, the cordycepic acid was the lowest among these three, about0.42times of that of Cordyceps sinensis and about0.5times of that of Cordyceps militaris fruiting bodies. The content of cordycepin in Silkworm Cordyceps worm body is higher than that of Cordyceps sinensis worm body, about2.66times of it. However, its polysaccharide and cordycepic acid content is lower than Cordyceps sinensis worm body, about0.38,0.05times of that of Cordyceps sinensis worm body, respectively. The cordycepin content of whole Silkworm Cordyceps cultivated was higher than that of Cordyceps sinensis, about2.6times of Cordyceps sinensis. While its polysaccharide and cordycepic acid content is lower than that of Cordyceps sinensis, about0.91and0.24times of Cordyceps sinensis. Amino acid content in Silkworm Cordyceps were higher than that of Cordyceps militaris fruiting bodies. The total amino acid content is about2.57times of Cordyceps militaris fruiting bodies, with essential amino acids of1.66times of Cordyceps militaris fruiting bodies, and half essential amino acids of about1.6times of Cordyceps militaris fruiting bodies.
