NAS multiple preparation is made up of a variety of natural wild herbs and its active ingredient is Hedyotis diffusa,Sedum sp. and Artemisia apiacea. It has been extracted from these herbs through fermenting and compounding vitamins in it. It has been confirmed that it can effectively prevent and cure the domestic animals and fowls diseases, especially in a certain range of concentration, highly pathogenic avian influenza virus can be killed up to 99.99%. In addition, NAS multiple preparation can heal large wound and make the injury diminish inflammation quickly, at the same time, it has obvious effects on respiratory system diseases.【Objective】In order to make an security evaluation of NAS multiple preparation and provide a theoretical basis for the clinical application, animal toxicology experiment was proceeded in this study.【Method】Animal toxicology experiment of NAS multiple preparation was divided into the acute toxicity test and the subacute toxicity test. On the basis of determineing absolute lethal dose(LD100)and maximal tolerance dose(LD0)of NAS multiple preparation on mice, the range of giving-dose was established and acute toxicity test was done to confirm the median lethal dose (LD50); then, subacute toxicity test was proceeded on chickens and rats to make sure the highest dose according to the range of 1/5~1/20LD50.【Results】(1)Observe mice in 6 giving-dose groups (4.8g/kg,6.4g/kg,8.5g/kg,11.4g/kg,15.2g/kg and 20.3g/kg) of pre-acute toxicity test of NAS multiple preparation every morning and evening for 7 days continuously, there were 1 mouse in 6.4g/kg dose group, 3 mice in 8.5g/kg dose group, 4 mice in 11.4g/kg dose group and 5 mice in 15.2g/kg and 20.3g/kg dose groups to die ,especially animals in this two dose groups died within 10 minutes after giving drugs, but all 5 mice in 4.8g/kg dose group lived. There were no abnormal changes on eating food, drinking water, spirit, activity conditions and dejecta compared with those in control groups. All these results confirmed that the absolute lethal dose(LD100)of NAS multiple preparation was less than 15.2g/kg and the maximal tolerance dose(LD0)was more than 4.8g/kg. (2)The acute toxicity test on mice determined that the median lethal dose(LD50)of NAS multiple preparation was 9.0573±0.0309g/kg and 95% confidence limit was 7.8777g/kg~10.4136g/kg. (3)There were no death in three chicken-dose-groups(0.9g/kg,0.18g/kg and 0.06g/kg)and three rat-dose-groups(0.7g/kg,0.18g/kg and 0.10g/kg)of the subacute toxicity test, their clinical behavior, body increasing had no abnormal changes after giving drugs for 14 days continuously compared with animals in control group; Physiological and biochemical parameters of blood of rats were normal on the 15th day; Killing all animals on the 16th day to do autopsy, observe histological and ultrastructure changes and determine organ coefficient, all the results were normal compared with the corresponding characters of animals in the control group.【Conclusion】The acute toxicity test on mice confirms that NAS multiple preparation belongs to low-toxic drugs and the results of the subacute toxicity test shows that this preparation is safe and no toxic side-effects on animals.
