The Pathological Studies On Subacute Toxieology Of Sudan Ⅰ In Mice

Sudan red,an unnatural pigment,is a kind of dyes to be used in chemical industry,and belongs to azo dyes.It is named to Sudanâ… -â…£according to their structure Sudanâ… and Sudanâ…¡belong to monoazo compound,while Sudanâ…¢and Sudanâ…£belong to bis-azo compound.Aniline and naphthalend,may be carcinogenic substance,are produced in the degradation of Sudan deys.In 1975 the International Agency for Research on Cancer(IARC),assessed Sudanâ… as a Group 3 carcinogen.In order to clarify the damage of the Sudanâ… on toxicology experiment which include acute, subacute test with Kunming mice.On the basis of determineing absolute lethal dose(LD100) and maximal tolerance dose(LD0) of Sudanâ… on mice,the range of giving-dose was established and acute toxicity test was done to confirm the median lethal dose(LD50);then,subacute toxicity test was proceeded on Kunming mice to make sure the highest dose according to the range of 1/7-1/28LD50.Moreover,this paper tried to probe into the effect mechanisms of its toxicity damage in liver, spleen,kideny,which provided the basis for the fully estimate of toxicology on mammals.The results showed:(1) Acute toxicity testThe absolute lethal dose(LD100) peroral on mouse of Sudanâ… was less than 6000mg/kg.bw and the maximal tolerance dose(LD0) was more than 1000mg/kg.bw.The median lethal dose(LD50) peroral on mouse is 2800.8233mg/kg.bw,95% confidence interval is 2200.887-3400.7596mg/kg.bw.According to the standard of acute toxic classification,Sudanâ… is low-toxic whose toxicity is the third-class.(2) Subacute toxicity testThere was cease and lose weight in the highest dosage group of male mice after exposure 20 days(P<0.05-P<0.01);And make no difference on weight of female mice.The Sudanâ… caused the decrease in the immune cell quantity and Hb quantity,which showed relationship between severity of the changes and increasing doses.Killing all animals on the 61th day to do autopsy,observe histological and ultrastructure changes and determine organ coefficient,the coeffcient of liver/body,spleen/body and kidney/body were increased in mice compared with the control group exposure to Sudanâ… after 60 days(P<0.05-P<0.01). At the same time,the organs were injured in different degree when pathological examination was made.Observation of ultrastructural and show minute points,verified that Sudanâ… can damage the mice liver,spleen,kidney and degrade physiological functions.As judge and analyze toxicosis effect,variance of pathology organization has offered sufficient morphology proof,after use different dosage of Sudanâ… .
