The study included two parts, lily tissue culture and lily virus-free. In the study of lilytissue culture, different organs(bulb scale, filament) of two lily cultivars, Lilium orientalis cv.Siberia and Lilium orientalis cv. Stargazer, were taken as explants, the best mediums fordifferentiation and enrichment were selected out by the orthogonal experiment, radication andtransplantation experiments were carried out at the same time. Lily bulbs were taken asmaterials for virus-free experiment, increasing heat treatment was used and tested by LSV tobe effective. The results showed as follows.1. The best lily bulb scale induction medium for Siberia and Stargazer was MS +6-BA2.0 mg/L + 2,4-D0.01 mg/L + NAA0.3 mg/L and MS + 6-BA2.0 mg/L + 2,4-D0.05mg/L + NAA0.3 mg/L respectively.2. The best lily bulb scale enrichment medium for Siberia and Stargazer was MS +6-BA2.0 mg/L + 2,4-D0.01 mg/L+ NAA0.5 mg/L and MS + 6-BA3.0 mg/L + 2,4-D0.03 mg/L+ NAA0.1 mg/L respectively.3. The best medium for inducing calli from filament and differentiation were MS +6-BA1.0 mg/L + IBA0.5 mg/L for Siberia and MS + 6-BA2.0 mg/L + IBA0.5 mg/L forStargazer.4. The best medium for enrichment of filament of Siberia and Stargazer was MS +6-BA2.0 mg/L + IBA0.1 mg/L + NAA0.3 mg/L and MS + 6-BA3.0 mg/L + IBA0.3 mg/L +NAA0.3 mg/L respectively.5. Soilless culture was the best for transplantation, and the survival rate was 100%.6. The best temperature of heat treatment for Siberia and Stargazer was 40℃-45℃.7. The effect of eliminating virus disease of increasing heat treatment of Siberia wasthrough; and Stargazer was general. The precentages of virus-free was 100%.
