Oriental lily is a perennial Liliaceae, lily bulbs like flowers, big beautiful, rich colors, rich flavor, has a high ornamental value and economic value, is one of the four in the world. In recent years, the disease led to serious decline of Oriental lily in quality, and constrained the development of Oriental lily. Oriental lily virus-free technology currently is prevailing operational difficulties and lower rates of detoxification, and therefore the establishment of user-friendly, virus-free rate of the virus detoxification system is to reduce the harm to Oriental lily and is resumption of the kind of one of the key issues.This paper study the optimized research of tissue culture technology of Oriental lily; and the research of higher temperature shoot tip culture technology of Oriental lily; and the Virus-free technical research. The main findings are as follows:Firstly, the optimized research of system of Oriental lily tissue culture: the results show that the best disinfection of Oriental lily stems and scales is 70% alcoholic immersing the best tedious site is bulblet lower induction of differentiation bulblet, induction rate is 100% and the number of induction by several small bulb is 10.5; it has a positive effect that add the activated carbon into induction by a small bulb of Oriental lily; Oriental lily bulb in the small-induced growth hormone NAA is better than IBA; Different concentrations of medium to small Oriental lily bulb-induced effects are very different, the better medium-induced is MS medium to small bulb of Oriental lily; they have a greater impact on 6-BA and NAA different concentrations and proportion of small bulb-induction of Oriental lily, the best medium of induction of Oriental lily bulb is MS+6-BA0.5mg/L +NAA0.30.5mg/L +AC3g/L; IBA on small Oriental lily bulb rejuvenation is superior to the NAA; 0~0.5cm level of the best small bulb and rejuvenation hormone concentration ratio is 6-BA0.1mg/L+IBA0.1mg/L, 0.5~1.0 cm, 1.0~1.5 cm and more than 1.5 cm level of the best small bulb and rejuvenation hormone concentration ratio are 6-BA0.3mg/L +IBA0.3mg/L; the best value-added training of Oriental lily is 6-BA0.7mg/L +NAA0.3mg/L; Aseptic material on the bulb once again divided the way of value-added effect of the Oriental lily compared to the conventional value-added effect of training(value-added coefficient is 9.5) increased more than double(20.5),the appropriate medium is MS + IBA0.3mg / L + AC3 g / L to rooting culture of Oriental lily . Secondly, the research of higher temperature shoot tip culture technology of Oriental lily: the results show that the normal maximum temperature is 32℃for shoot tip growth of Oriental lily; high concentrations of MS medium is more suitable for the shoot tip culture of Oriental lily; IBA is more suited to the lily shoot tip culture under 32℃; the ideal medium of shoot tip culture of Oriental Lily is MS+6-BA0.1mg/L+IBA0.1mg/L+AC3g/L, Its effect is better than MS +6-BA0.05mg/L+IBA0.05mg/L;Dark shoot tip culture of Oriental lily did not affecteThirdly, the Virus-free technical research: the results show that three methods could emerge the some cucumber mosaic virus and Lily of the virus, the Worst effect is chemical and shoot tip culture, two viruses emerge all the ratio was only 26.7%; heat treatment and shoot tip culture in the two viruses are extrusion rate was 41.7%; the best effect is shoot-tip under high temperature and several times with the long-term culture, virus-free rate is 100%. Shoot tip that a combination of several high-temperature training subculture detoxification programme is superior to combination of heat treatment shoot tip culture and chemical processing of the shoot tip culture, On the one hand, a higher rate of virus-free is up to 100%, On the other hand, shoot tip's material is larger and easier operation. Higher temperatures shoot tip culture with several subculture detoxification study also found that simply shoot tip culture play a minor role in the cucumber mosaic virus and Lily of the virus, with the subculture of the increase in the number of virus-free rate substantially improve, After five detoxification treatment (one shoot tip culture and four subculture of the virus-free treatment), virus-free rate is 100%. The results also show that the cucumber mosaic virus and Lily of the virus more easily emerge.The experimental study of shoot tip culture in higher temperature add multiple oriental lily virus-free scheme possess below two large advantages: firstly, he length of shoot tip exceed 1 mm, virus free operating completely can depend on eye below progressive ,and the survival rate of shoot tip obviously improve; secondly, the virus free rate enormously enhance, the vims free rate can reach on 100% when virus-free treat, by five times. If our operation is skilled, the seeding by five times virus-free treatment may enter the production directly and does not need to examine. This may save the expensive examination reagent and greatly reduces the production cost of the virus-free seedling. Therefore, we thought that this method has the significant significance for the research and production of the virus-free seedling of the Oriental lily.
