Houuuynia cordate Thunb.the whole plant of Houttuynia cordata Thunb.,is a kind of traditional Chinese medicine and also a kind of delicious vegetable.It is the raw material resources of Yuxingcao injecting liquid.In current time,pharmacological experiments had proved that it had been identified as one of a kind of most potential resources which could be used as both medicine and food by National Health Department.On the one hand this study through plant tissue culture technology establishment Houuuynia cordate Thunb.regeneration system,on the other hand take the volatile oil,the methyln-nonylketone,the total flavanone,the chlorogenic acid,the quercetin,the amino acid,the mineral substance,the Vitamin,the rough fat,the soluble total sugar the quality synthetic evaluation target,conduct the comparison research through the chemical constitutents content in different popupations of Houuuynia cordate Thunb.for the heredity resources protection,the improved introducing new species and the use of Houuuynia cordate Thunb.provides the theory basis.1.Establishment of regeneration system in Houuuynia cordate Thunb.This study slected the most suitable culture medium and the best cultivating explant for Houuuynia cordate Thunb.regenerated planted by the indeterminate bud and somatic embrygensis approach,and an efficient in vitro plant regeneration in Houuuynia cordate Thunb.was established.The indeterminate bud of Houuuynia cordate Thunb.inducing best medium was MS +6-BA1.5 mg/L + NAA 0.1 mg/L;the bud was pointed to be the most suitable explant. The indeterminate bud proliferates the best medium was MS+6-BA 2.0 mg/L+2,4-D0.1 mg/L+NAA 0.1 mg/L;The best rooting medium was MS+IBA1.0mg/L;The most suitable sugar concentration was 30 g/L,the most suitable pH was 5.8.The orthogonal test result indicated that the best medium for inducting embryogenic callus from leaf blade of Houuuynia cordate Thunb.was modified MS+2,4-D 1.0 mg/L +6-BA0.5 mg/L;the best medium for inducting loosen callus was modified MS+2,-D2.0 mg/L+6-BA0.5 mg/L.Both can inducting embryogenic callus take out somatic embryogebnesis further,and may induce the leaf blade and the petiole to produce somatic embryogebnesis directly on the modified MS+6-BA 1.0 mg/L solid medium.The best transplanting technique was also obtained,which was taking"frog stone + perliteten + nutrition soil"(1:1:1)as matrix,the transplanting survival percent was ever 96%using this technique. 2.The cytology observation results showed that chromosome mumbers of Houuuynia cordate Thunb.was 20,22,24,26,30,32,of which the number of 20,26,30 and 32 were found for the first time.So,the chromosome mumberand karyotypic were different among diffrent populations.3.The comparative anatomy.observation result showed that there were significant differences in the root,subterraneous stem,aerial stem,leaf and so on.From the results of this study:The volatile oil in Houuuynia cordate Thunb.was related to the quantity of oil cell,numbers of Vascular bundle and arranges of palisade tissue in subterraneous stem,aerial stem,and leaf.The content of volatile oil in Houuuynia cordate Thunb.was higher,when the quantity of oil cell and vascular bundle increased,and the palisade tissue arraied relatively loose.4.The analysis on medical constitutents result that there were significant differences in the contents of the volatile oil,the methyl nonyl ketone,the total flavanone,the chlorogenic acid,the quercetin.The contents of volatile oil from Houuuynia cordate Thunb.were obtained with steam distillation.The result showed that the content of volatile oil in Chaohu was greatest 0.229%,the contents in Huangshan and Hefei came next,0.162%and 0.142%, respectively,the content in Hubei was least 0.082%only.The contents of methyl nonyl ketone from Houuuynia cordate Thunb.were deter mined by GC.The result showed that the content of methyl nonyl ketone in Huoshan was greatest 0.102 mg/g,the content of Guangde and Chaohu came next,0.093 mg/g and 0.094 mg/g,respectively,the content of Tong ling was least 0.015 mg/g only.The contents of total flavanone from Houuuynia cordate Thunb.were determined by Ultraviolet differential sectorphotometirc method.The result showed the content of total flavanone in Huangshan was greatest 13.26 mg/g,the content of cultivar plant came next, 11.84 mg/g,the content of Tong ling,Anqing and Shi tai contents of total flavanone also comparatively highly,0.62 mg/g,10.61 mg/g,10.04 mg/g,respectively,the content of Lu zhou contents was least 4.37 mg/g.The contents of the chlorogenic acid and quercetin.from Houuuynia cordate Thunb. were determined by HPCE.The result showed the content of chlorogenic acid from Chaohu was greatest 3.44 mg/g,the content of Tong ling came next,3.05 mg/g,the content of Neijiang was least 0.31 mg/g;the content of quercetin from cultivar plant was greatest 7.26 mg/g,the content of Chaohu came next,5.14 mg/g,the content of Shitai was least 1.17mg/g. Making reference to the chemical constitutents comparative result,mark the integrat ed evaluation different populations of Houuuynia cordate Thunb.The conclusion is as follow:The Houuuynia cordate Thunb.collected from Chaohu,has higher medical value, and quality had better synthetically in all above index valuation.It was the best ermplasm for introduction.5.The analysis on nutrition constitutents result that there were significant differences in the concent of amino acid,mineral elements,Vitamin,soluble sugar and rough fat.The determination of amino acid showed that the total concent of the amino acid in Ji xi was greatest 2186.13 mg/kg,the concent of Shitai came next,1108.23 mg/kg,the concent in Huoshan was least 400.71 mg/kg.The determination of mineral elements showed that The contents of K,and Ca mineral elements were higher than other elements.The contents of K was greatest in Anqing,6243 mg/kg;The contents of Ca was greatest in Hefei,1237 mg/kg;The contents of Fe was greatest in Huangshan,1289 mg/kg;The content of Fe was least in cultivar plant,6243 mg/kg;The contents of Zn was greatest in Huoshan,8.0mg/kg;Pb and Cd were not be found in all the populationsThe determination of Vitamin showed that the total concent of Vitamin in Jixi was greatest 396.59μg/g mg/kg,the concent of Vc in cultivar plant was greatest 36.4μg/g.The determination of soluble sugar showed that the concent of soluble sugar in Guangde was greatest 3.91%,the concent of Tongling came next,3.82%,the concent of Zigong was least 1.07%.The determination of rough fat showed that the concent of rough fat in Dujiangyan of sichuan was greatest 7.82%,the concent of Chuzhou came next,7.22%,the concent of Chaohu was least 2.35%.Making reference to the nutrition constitutents comparative result,mark the integrated evaluation different populatins,of Houuuynia cordate Thunb.,The conclusion is as follow: The Houuuynia cordate Thunb.collected from Jixi,having the nutrition component rich and higher nutrition value for introduction..
