Using tubers of Pinellia ternata of 6 different populations-Xieqiao Tongren,Zhaihao Rongjiang,Sandu Zunyi,Huaxi Guiyang, Xinlongchang Xingren and Yemachuan Hezheng-in Guizhou Province as explants,rapid propagation systems of one-step-seedling formation culture of excised tubers were established via comparing effects of different phytohormones' combinations for the culture of excised tubers of these 6 populations,and the phytohormones' combinations which were fitted for all of the 6 populations were screened out,providing some experimental references for multiple production of seedlings of different populations under the same tissue culture conditions.The contents ofβ-sitosterol,total alkaloid,total protein and guanosine were determined and the experiment of pharmacodynamics of P.ternata in those 6 populations was conducted, in order to providing some preliminary references for rapid propagation,artificial plant and further researches,which would be based on the comparison and assessment of the quality of ternata.The main experimental results were as following:1.Regeneration systems of P.ternata in the 6 populations were established from tuber explants.The effects of disinfector combination on the disinfection of tubers were studied.The results showed that the optimal disinfector combination was 75%ethanol 30s-2%NaClO 3min-0.2%HgCl210min.This treatment could obtain about 82.5%sterilized explants,which has been primarily employed in explant disinfection of micropropagation of tubers from the 6 populations in vitro.When tuber segments were cultured on MS medium supplemented with proper 2,4-D and 6-BA,NAA and 6-BA,NAA and KT,plants were regenerated directly.The MS medium added with 0.5mg·L-1 2,4-D+1.0mg·L-16-BA,0.5mg·L-12,4-D+1.5mg·L-1 6-BA,1.0mg·L-1NAA+1.5mg·L-16-BA were fitted for one-step-seedling formation culture of all the 6 populations.Cultured on MS medium supplemented with the 3 phytohormones' combinations,per explants possessing 6.50~12.00 differential seedlings.After 60~64 days of culturing,plants were regenerated directly.15 days after transplanting,the average survival rate was above 90%.2.Comparison study on 4 chemical components of the six different populationsβ-sitosterol,total alkaloid,total protein and guanosine were tested,whose contents would be the criterion to assess the quality of P.ternata in different populations.The results showed that contents of the 4 chemical components in the 6 populations were different.The highest contents ofβ-sitosterol and total protein appeared in Xinlongchang Xingren population,the content of total alkaloid in Xieqiao Tongren population was the highest one,the guanosine content of Huaxi Guiyang population was higher than.the others.The result of Paired-Samples T Test indicated that the diversity of them were no significant prominent.All of the six populations could be classified into four types through Hierarchical Cluster Analysis.The populations of Sandu Zunyi,Huaxi Guiyang and Yemachuan Hezheng were classified into one type,while Zhaihao Rongjiang,Xieqiao Tongren,Xinlongchang Xingren were classified into three types.3.Comparison study on pharmacodynamics of the 6 different populationsThe diversity of pharmacodynamics of P.ternata in different populations was expedited by antitussive effects of the low-dose and high-dose water extract.The results showed that both the low-dose and high-dose water extract of the 6 populations demonstrated different antitussive effects for mice' cough.The result of Regression Analyze showed that the linear relationship between the time of administration and the frequency of coughing in 1 min,1 hour after administration,was obviously prominent.According to the regressive equation,the antitussive effect of high-dose was better than the low-dose ones.The antitussive effect of high-dose water extract of P.ternata displayed:Yemachuan Hezheng>Huaxi Guiyang >Sandu Zunyi>Xieqiao Tongren>Zhaihao Rongjiang>Xinlongchang Xingren.
