| The study is on 32 Xishan's Series maize hybrids and inbred lines popularized of by SSR molecular markers and their application in Guizhou. There are mainly three aspects discussions on system of PCR amplification,Electrophoresis optimized and construction,analysis and application of SSR fingerprint Models. The results are as follows:(1) The affecting factors of PCR amplification: There are no differed in DNA from maize germ and leaves, small affection from DNA board,TaqE and primer's capacity, large affection from Mg2+ and dNTP levels in the system of PCR amplification.(2) Electrophoresis optimization: When we practice on Electrophoresis, we may shorten pre- Electrophoresis's time to 3 minutes, may also drop voltage and gum concentration for saving time.(3)Fingerprint model's construction: The fingerprint model's construction is by main electrophoresis banding pattern between 100bp and 200bp which is similar with the rice. We attain 22 core primers from 100 SSR primers, three of which may distinct all maize inbred lines, three of which may distinct all maize hybrids lines.(4) electrophoresis banding patter's analysis: We attain four types from our lab which are accordant,complemented,leaning and mixed models by main electrophoresis banding pattern between maize hybrid and inbred lines. And there were remarkable changes on assisted electrophoresis banding pattern in maize inbred and hybrid lines.(5)Maize finger model's application: SSR molecular markers may be used to the research of maize genetic diversity and seed purity's appraisal. And we attain four results from our experiment. Firstly, we should introduce the germless of highland and cold region into our area. Secondly, we should further utilize the inbred maize of Xi1-2000-2415 and Su-37, and have an eye to breeding high qualities maize inbred. Thirdly, There are mainly hybridized model by the germless of tropical and subtropical zone in the maize of Xishan series. Finally, hybrid maize is not positive correlation with genetic distance. |
