Prolactin is an important gonadotropin, which regulate the activities of animals, especially with the growth and reproduction of animals. It play an important role in mammalian lactation, litter and production performance of poultry. The bone morphogenetic proteins (BMPs) are members of the transforming growth factorβ(TGFβ) superfamily. Which is extensively involved in animal embryonic development, the immune system, reproductive and developmental follicle development. BMPs cannot exert their physiological functions without bone morphogenetic protein receptors. Therefore, the study of BMPR is of great significance to animal embryonic development, growth, reproduction ,polymorphism of PRL gene and BMPR-IB gene is topic of this article. To explore the relationship between two candidate genes and the high productivity of Jining grey goat.The purposes of this study were to detect single nucleotide polymorphisms (SNPs) within PRL gene and BMPR-IB gene, to analyze the genetic structure of three goat breeds, and to investigate the effects of the two genes on litter size of Jining Grey goats. Based on the bovine and ovine DNA sequences in GenBank, five and ten primer pairs for PRL gene and BMPR-IB gene, were designed for PCR respectively. Amplified regions included five exons of PRL gene, and exon 1, exon 3, exon 6 to exon 10 and 3′untranslated region (UTR) of BMPR-IB gene. In both high fecundity breeds (Jining Grey and Wendeng dairy goat) and low fecundity breed (Inner Mongolia Cashmere goat) by PCR-SSCP The genetic structure of the three populations at polymorphic loci, and the associations of the litter size of Jining Grey does with the two genes were also explored. The results are as follows:(1) There was no single nucleotide polymorphism on the PRL gene in the four goat breeds.(2) The PCR products of two pairs of primers were polymorphic among those of ten pairs of primers. polymorphic loci B8 and B10 were on 3′untranslated region (UTR).(3) At locus B8, three genotypes (AA, AB and BB) were detected in Jining Grey goats and Wendeng dairy goats, two genotypes (AA and AB) were detected in Inner Mongolia Cashmere goats. Sequencing revealed one mutation (71C→T) of BMPR-IB gene in the genotype BB in comparison to the genotype AA. At locus B10, three genotypes (CC, CD and DD) were detected in Jining Grey goats and Wendeng dairy goats, one genotype (CC) was detected in Inner Mongolia Cashmere goats. Sequencing revealed one mutation (130T→C) of BMPR-IB gene in the genotype DD in comparison to the genotype CC.(4) By running a popular analysis tool, We studied the differences of LSM for litter size. There were non-significant (P>0.05) in three genotype of Jining Grey goats.(5) Because of the different experimental methods, some of which may be mutation was not detected, so we can not rule out the possibility that these two genes are the main influence goats high fecundity gene.
