Faba bean (Vicia faba L.) is one of the important grain legume crops in the temperate and subtropics. The objective of this work is to assess the genetic diversity of autumu-sown faba bean germplasm, as well as the relationships between accessions and their geographic origins, and the way of core collection form, in order to generate useful information for collection, conservation and usage of winter faba bean genetic resources.AFLP finger printing system was optimized by employment of 6 faba bean accessions of geographic difference, for genetic diversity analysis. 10 pairs of informative primers were screened and used for AFLP analysis to reveal genetic diversity of faba bean.243 faba bean accessions genetic resources were analyzed by amplified fragment length polymorphism (AFLP) methodology, using the 10 informative primer pairs. In total, 323 unambiguous bands were created, of which 266 were polymorphic, covered 82.4% in total. According to the dendrogram by UPGMA cluster analysis using SM similarity coefficient calculations based on AFLP banding, all the accessions were distinct and revealed enough genetic diversity for identification and classification of accessions within tested faba bean genepool.The results of the analysis on morphological characters showed that winter faba bean were unsensitive to different ecological conditions. The average Shannon-Weaver index was 0.8661, the Shannon-Weaver index of seed shape and seed coat color are greater than the aveage, and the Shannon-Weaver index of hilum color is the smallest. Among provinces, the index of landraces from Guizhou province is biggest. Based on morphological data, 243 accessions were clustered into 2 groups on 0.644 similarity level, Yunnan province accessions were clustered into one distinct group.The PCO and cluster analysis based on AFLP and morphological data showed that accessions from Yunnan province differ to the accessions from other provinces, several accessions from Sichuan, Guizhou and Zhejiang provinces are distinguishing. Faba bean accessions from Yunnan, Sichuan, Guizhou and Zhejiang should be enhanced in the genetic resources work on collection, evaluation and conservation.The methodology for the the construction of core collection based on combination of morphological and AFLP markers is the best to those by using morphological marker and AFLP marker alone.
