| A study was conducted to seek an efficient and environmental mycotoxin organic adsorbent. This experiment investigated the extraction technique of glucomannan and adsorption effect of aflatoxin by using the discarded yeast in brewery. including two experiments.In experiment 1 , glucomannan was extracted from the discarded yeast in brewery by Na2HPO4-citric acid buffer solution. The optimal conditions was buffer solution at pH 7.0, 120°C, 90min as well as 2 times extraction. Extract of yeast was deproteinized by Sevage. The polysaccharide was white, flavourless and soluble in water and insoluble in organic solvents, with 90.07% of polysaccharide, 88.25% of glucomannan and 2.53% of protein.In experiment 2, one hundred and eighty 8-day-old AA broilers were equally divided into three groups (six replicates per group with ten broilers per replicate), three dietary treatments of control feed without addition of glucomannan and aflatoxinB1, 100ppb AFB1, 100ppb AFB1+0.1%glucomannan were fed to treatment groups for twenty-one days. Six broilers/group were slaughtered at 7, 14, 21, 28 days of treatment. Body weight(BW) and feed consumption were recorded to calculate average daily gain(ADG), average daily feed intake(ADFI) and feed/gain(F/G) for each replicate. Serum total protein, albumin and transaminase were measured. Indexes of liver, kidney, pancreas, heart, spleen, thymus, bursa were recorded. Hepatic enzymes were measured. Main results were obtained as follow:1. Growth performance of broiler was affected(P<0.05) by 0.1mg/kg AFB1. BW, ADG, ADFI were depressed significantly. F/G was increased (P>0.05). The content of Serum total protein and albumin were decreased (P<0.05) and activity of serum glutamic-pyruvic transaminase and glutamic-oxalacetic transaminase were increased (P<0.05) significantly. Weights of liver, kidney, spleen, heart were increased (P<0.05) significantly and weights of thymus, bursa were reduced. Hepatic succinate dehydrogenase and cholinesterase were decreased(P<0.05), alkaline phosphatase,γ-glutamyl transpeptidase were increased(P<0.05), and glutathione peroxidase and superoxide dismutase were decreased. Hepatic malondialdehyde content was decreased. The histopathological liver changes included: tumefaction steatosis necrosis and the ductular proliferation. 2. Related parameters was returned to normal level by the addition of 0.1% glucomannan to AFB1-contaminated diet, showing beneficial effects of glucomannan on aflatoxicosis.These findings indicated that glucomannan had ability to recover the AF-decreased performance, organ damage and to correct aberrations in related parameters and glucomanan can effectively diminish the deleterious effects of AFB1 in AA broiler.and glucomannan would be a potential ameliorator of aflatoxicosis in broiler. |
