β-agonist is one kind of veterinary. They can enter human body through food. They may lead to food poison and cause harm to human health when they exceed a certain quantity. Therefore, the detection ofβ-agonist residue has been strengthened in all countries of the world.In this paper, on the background of lack of analysis method forβ-agonist residue in animal-derived food, an analysis method forβ-agonist residue in liver and muscle tissues was investigated systematically. The pretreatment method of sample was discussed and the simultaneous detection of fiveβ-agonists using an LC-MS method has been established.During the pretreatment process, extraction ofβ-agonists in sample was carried out by liquid-solid extraction and the extract was cleaned up by C18 SPE cartridges. The effects of extraction solvents and extractions times were investigated and the best extraction conditions have been established. The optimal conditions of solid-phase extraction were also studied. Average recoveries of spiked analytes ranged from 77.2% to 95.6% with low RSD of 1.9% to 8.3%.The conditions of LC-MS were developed for simultaneous detection of fiveβ-agonists in liver and muscle tissues. Under certain conditions,β-agonists could be separated and RSD were ranged from 0.5% to 1.4%. The limits of detection for the method could be reached at 0.2-0.5 ng/g.The analysis of the spiked samples showed that the established pretreatment and LC-MS methods could be applied to analysisβ-agonist residue in these tissues. The results have highly reliability.
