Powdery mildew, caused by Erysiphe.graminis f.sp.Ttritici, is a destructive disease of common wheat throughout the world. In view of high genetic variability of Erysiphe.graminis f.sp.Tritici, identifying resistant genes and using it to breed resistant cultivars is the most economical and effective approach to control this disease. Therefore the exploitation of Powdery mildew resistant genes, which included its marking, location and molecular marker aided selection, would benefit breeding the effective Powdery mildew resistant cultivars. In this study, the Powdery mildew resistant gene of variety YanZhan2Hao was analyzed by using SSR technique. The results are showed just as follows:1. Cross was carried out between YanZhan2Hao as resistant parent and CangMai030 as susceptible parent, and F1 plants from the cross were selfed to obtain F2 population. In F1 population, every individual had resistance to Powdery mildew. But F2 population was divided into two kinds: resistant individual and susceptive individual, the ratio between resistant individual and susceptive individual accorded with 3:1. Genetic analysis concluded that the resistance was controlled by a pair of single dominant gene.2. Bulked segregant analysis (BSA) was used to establish resistant and susceptive gene pool in order to search for SSR markers with polymorphism. The PCR results amplified by SSR primers of these F2 individuals were analyzed by using Mapmaker/EXP (VERTION 3.0b) and Map manager. The results indicated that this gene was located on the chromosome 2D.3. The genetic map which included resistant gene to Powdery mildew of YanZhan2Hao and three SSR markers was constructed.
