Swine fever is a strong contact with sexually transmitted diseases of pig, it was listed by the world animal health organization in OIE disease list and responsible for a large number of economic loss to pig industry, the swine fever was the first kind of infectious disease in our country.In recent years, studies have shown that yupinfeng powder, and with gamma globulin in traditional Chinese medicine (TCM), was a strong Chinese medicine immune enhancer. Chinese traditional medicine superfine grinding technology used modern technology to traditional Chinese medicine or Chinese medicine extract superfine pulverization, can make effective to improve at the same time greatly reduce side effects, so the Chinese traditional medicine superfine pulverization has the vital significance.This study will make yupinfeng powder into superfine powder preparation, and its physical properties are studied, using the HPLC determination to detected the prim-O-glucosylcimifugin of superfine powder, study of its impact on swine fever antibody levels in the last.Experiment to select good quality of Astragalus membranaceus (Fisch.) Bunge, Atractylodes macrocephala Koidz and Saposhnikovia divaricata (Trucz.) Schischk. wash, dry, respectively prepared Chinese medicine coarse powder first, and then take a variety of coarse powder superfine grinding to the superfine powder. Take a variety of samples by using relevant instruments for particle size distribution, specific surface area, liquidity and hygroscopic. Particle size distribution and specific surface area, according to the results shows that particle size distribution of coarse powder was in 50-100 μm range and superfine powder was in 1-10μm range, specific surface area of the superfine powder was times larger than that of coarse powder. According to the results of superfine powder and coarse powder of Angle of repose ratio greater than 1, shows that the superfine powder liquidity decreases. Hygroscopic results shows that, under the condition of temperature 25℃, relative humidity 75%, Astragalus membranaceus (Fisch.) Bunge coarse powder, Astragalus membranaceus (Fisch.) Bunge superfine powder and Atractylodes macrocephala Koidz superfine powder are basic balanced moisture absorption at 96 h. and Atractylodes macrocephala Koidz coarse powder, Saposhnikovia divaricata (Trucz.) Schischk coarse powder and Saposhnikovia divaricata (Trucz.) Schischk superfine powder are basic balanced moisture absorption at 120 h.HPLC was used for the preparation of superfine powder for prim-O-glucosylcimifugin in determination of detection wavelength of 254 nm, mobile phase of acetonitrile-water (30:70). The concentration of 100.0μg/mL,50.0μg/mL, 25.0μg/mL,12.5μg/mL,6.3μg/mL of reference substance solution respectively from 20 μL injected into HPLC, with peak area as the ordinate, the sample quantity as the abscissa denotes, drawing standard curve. Then carries on the precision, repeatability and recovery experiment, content determination for the last three batch samples. HPLC figure, according to the results of prim-O-glucosylcimifugin reference substance around the retention time of 9.2 min, and the sample at about 9.2 min also appears a good peak shape, smooth and baseline. Standard curve regression equation obtained for Y=8879.65 X-7823.36, r=0.99995 (n=5), the test shows that prim-O-glucosylcimifugin reference substance had good linear relationship in the range of 6.3-100.0μg/mL, and the minimum detectable was amount 0.46 μg/mL. Experimental results show that the precision of prim-O-glucosylcimifugin element reference substance peak area integral value, RSD was 0.92%, the precision of this method shows better; Reproducibility, according to the results of sample prim-O-glucosylcimifugin element glycosides RSD was 0.89%, showed good reproducibility; The recovery experiment results show that the recovery rate of 97.52%, RSD was 0.95%. Content results show that the three batch were 0.82%,0.75%,0.79%, than some related yupinfeng coarse powder.Experiment selects the age of 65d, the ternary hybrid (D* L* Y) 50 heads of weaning piglets, average weight 30 kg, and injected swine fever cells miao head 2 per head, were randomly divided into 5 groups:group Ⅰ base diet group and 1% yupinfeng superfine powder; Group Ⅱ based diet plus 0.5% yupinfeng superfine powder; Group III base diet plus 0.1% yupinfeng superfine powder; IV group based diet plus 2% yupinfeng powder; V based diet group. By swine fever immune after 7 d,14 d and 21 d,28 d to test the antibody levels. Results show that the 7 d after immunization, swine fever antibody levels began to rise gradually,21 d after immune to reach the highest level, and then a slight decline in. Results show the swine fever antibody levels of the group with add 1% yupinfeng superfine powder group, adding 0.5% yupinfeng superfine powder, adding 0.1% yupinfeng superfine powder group and add 2% yupinfeng coarse powder were significantly higher than that of control group (P<0.05); Add 1% yupinfeng superfine powder group, adding 0.5% yupinfeng superfine powder group and adding 0.1% yupinfeng superfine powder swine fever antibody level were significantly higher than adding 2% yupinfeng powder meal group (P<0.05); Add 0.5% yupinfeng superfine powder group of swine fever antibody level was significantly higher than adding 1% yupinfeng superfine powder group and add 0.1% yupinfeng superfine powder group (P<0.05); Add 1% yupinfeng superfine powder group antibody levels higher than adding 0.1% yupinfeng superfine powder group, but no significant difference (P> 0.05). Results show that the yupinfeng superfine powder was immune to swine fever has significantly enhanced effect, and add 0.5% was the best.The experimental results show that each Chinese medicine of yupinfeng after superfine,the pulverization size and specific surface area increases, the liquidity decreases, and hygroscopic was stronger. HPLC figure, precision and recovery experiments show that this method was feasible. Prim-O-glucosylcimifugin element contents, according to the results of the results of the three batch were 0.82%,0.75%, 0.79%, than some related yupinfeng coarse poeder. Swine fever antibody testing results show that the yupinfeng superfine powder and yupinfeng coarse powder can significantly enhance the immune function of pigs, raise pigs swine fever antibody levels, however,the yupinfeng superfine powder was better than coarse powder, and add 0.5% was the best.
