Insects of the order Lepidoptera are one of the most serious pests of cereal crops inChina and around the world. Chilo supperssalis (Walker) and Chilo hyrax (Bleszynski)belong to the same order: Lepidoptera and family: Pyralidae. They have close semblancesuch that it is usually hard to differentiate them. C. suppressalis is an important pest ofrice plant while, C. hyrax is a major pest in reed fields. In this study, I investigated thegenetic diversity of C. supperssalis and C. hyrax using biochemical and moleculartechniques.At the biochemical level,we invested the detoxification enzyme and detoxificationenzyme.(CarE, TchE, ADH, SOD, CAT, POD) The CarE and the TchE in Chilo supperssalishas a higher level than in Chilo hyrax Bleszynski.And they have a visible difference.The resulesshowed that: in 3rd and 4rd,the peroxidase of Chilo supperssalis is lower than that of Chilohyrax Bleszynski. Oppositely in 5rd,which showed a visible difference .The protection enzymesuch as catalase and alcohol dehydrogenase showed a higher activity in Chilo supperssalis andhas a obviously difference between them. Oppositely the superoxide dismutase in Chilosupperssali is lower than that of Chilo hyrax Bleszynski.The protein types in these two insects also showed obvious differences in number,position, breadth and definition. And their molecular weight ranges from 0 to 97400Dalton. The C. supperssalis from different geographic region has similar protein types.But result of PAGE showed that the esterase isoenzymes and LDH isoenzymes in C.supperssalis and C. hyrax have some differences in types and intensity. Through clustersanalysis, I found that they can completely separate from the individuals, and their genedistance D is 0-0.64355 and 0-0.28870.At the molecule level,we used mt-DNA sequence analysis to research the two genes ofmitochondria. The results showed that :the ITS gen in Chilo supperssalis and Chilo hyrax Bleszynskihas 579 and 583 bp separately. There are 97 pieces of mutation between the two ITS gene sequence,and take 16.8% of total nucleoside. The 12sRNA gen in Chilo supperssalis and Chilo hyrax Bleszynskihas 399 bp pieces separately. There are 17 pieces of mutation between the two 12sRNA genesequence, and take 4.3% of total nucleoside.The results showed there are very near blood relations onthe development of the system between Chilo supperssalis and Chilo hyrax Bleszynski.
