| Selenium is a necessary trace element in human bodies. As a part of life activity, Selenium in human body has an important physiology function . It participates the human body metabolism, promotes the human body growth .As a necessary trace element, the lack of selenium and excessiveness of it have a very narrow scope , the lack and excessiveness lead to very serious aftereffect. The content in soil affects the content level of selenium in plant derectly, as selenium in the soil can deliver the vegetative chain by the plant, the selenium level in the soil affects the selenium in the plant and affects the health of human and animal directly . Some plants which have particular gene can enrich selenium of soil and the level of selenium is higher than other plant in the same condition . As raw material ,wheat is important source of food,selenium level in the wheat affects daily absorbability of people directly. The exact determination for selenium level in food has important meaning for disease and toxicity research. Colour wheat is a new breed ,selenium content level has not reported until now. The research offered a gist and reference for development this special function and has an important meaning for development district economy.It mainly includes the following five parts:1. Pretreatment of sample and choice of determination methodThe Microwave Digestion technology was a new method for pretreatment of sample .This method avoided pollution of environment and saved reagent. The digestion of wheat was 25min and the digestion of soil was 30min .The text explored the accession quantity of acid for effect of digestion .The wheat adopted 4.00mL HNO3 and 2.00 mL H2O2 and the soil adopted 2+4+1 HNO3 -HF -HClO4 7mL,these systems had better digestion effect. The text explored two methods for determination of selenium content- Ultraviolet Spectrophotometric Method and Fluorometric Method. By Ultraviolet Spectrophotometric Method ,the analytical results showed that selenium content was higher than Atomic Fluorescence Spectrometry,this method was not advisable for trace element,it had a system error .There was no difference between Fluorometric Method and Atomic Fluorescence Spectrometry, so we adopted Fluorometric Method for determination. The detection limit was 3.6×10-2ng/mL. the recovery was in range 95.64-103.69%,The analytical result was satisfactory.2. Deternination of Selenium content in six varieties wheats background and difference between varietiesBy Microwave Digestion pretreatment and Fluorometric, author determinated different parts of wheat ,containing whole wheat flour , cortex of wheat and center of wheat and distinguish with the selenium content in different part and difference of selenium content between six varieties wheats. In six varieties wheats background ,the result showed that the whole wheat flour was in rang 0.0643-0.136mg/kg ,cortex of wheat was in rang 0.100-0.155mg/kg, center of wheat was in rang. 0.0553-0.116mg/kg. Cortex of wheat had higher selenium content and center of wheat had lower selenium content in six varietyies wheats . The result was that selenium content of Common Violet Wheat was topmost, secondly was Horniness Violet Wheat, Gray Wheat and Blue Wheat was lowest. There was a notable difference for selenium content between four breed colour wheats and no notable difference between common wheats.3. Deternination of Selenium content in six varieties wheats of five area and difference between areaBy Microwave Digestion pretreatment and Fluorometric, author determinated selenium content of six varieties wheats in five area ,The result showed that selenium content of wheat in ShangQiu was topmost and JiaoZuo was lowest.,in rang 0.070-0.194mg/kg .The wheat in ShangQiu was notable difference with other area and background (seed),besides blue wheat ,background was notable difference same kind wheat what was planted in five area and was lower than the wheat what was planted in five area.4. Deternination of selenium content of soil in five area and differenceBy Microwave Digestion pretreatment and Fluorometric, author derminated selenium content of different time in Shangqiu,Luohe,Gongyi ,Zhengzhou,,Jiaozuo. The result showed selenium content of soil was topmost in rang 0.598-0.801 mg/kg before seedtime and was lowest in rang 0.532-0.712 mg/kg at harvest. There was a notable difference for Selenium content in soil between five area before seedtime and after harvest , there was no notable difference between Gongyi and Luohe in growth time , and there was notable difference between other area.5. Correlation of Selenium content between wheat and soil and Biological Asorptanc Author determinated selenium content of wheat and soil in five area and compared difference of selenium content in wheat and soil among five area and analized correlation between selenium content of wheat and soil. The result showed content of Zhou Wheat 16 was plus liner Correlative with selenium content of soil of three times (P<0.05) . Horniness Violet Wheat,Common Violet Wheat, Gray Wheat, Blue Wheat and YanZhan 4110 wheat were strong plus liner correlative with selenium content of soil of three times (P<0.01). Selenium content of the wheat increased with selenium content of the soil. In same condition ,there was different biological asorptance between wheat, Gray Wheat was lowest in rang 12.35-15.87%, Common Violet Wheat was topmost in rang 23.09-27.25%.
