The Hybrid F1 between Leymus chinensis and Leymus cinereus were used for the study materials. By study and filtration the best treat of F1 young spikes , hormone assored proportion of callus induction , cell suspension culture , sub-culture and differentiation culture of mature , which developed the system of plant regeneration ; Studies on chromosome doubling by colchicines and trifluralin liquid treating callus , suspension cells ,discussing the treating effect with different time and concention of different revulsant .The main results were as fellow:(1) The study induced F1 young spikes between Leymus chinensis and Leymus cinereus to produce callus , expants were cut up to culture ,which not only favored callus produce ,simultaneity , shortened time of producing callus and advanced examination efficiency .(2) F1 young spikes were cultured on MS containing 2mg/L2,4-D to induce callus , callus were better embryo and more differentiated .And free-hormone MS was the best for plant regeneration .(3) Callus were compact , relax ,crisp and graininess on the medium of 3mg/L2,4-D+MS . Result indicated which were aptly cultured in a shaking mode .(4) The study cultured Hybrid F1 between Leymus chinensis and Leymus cinereus to develop the cell lines of suspension , result indicated that 4%sucrose was the best .In the suspension culture ,it was key that callus was transferred from solid medium to liquid medium ,every sub-cultured 1/3 original medium was come out .In this way ,cells developed quickly and enriched contain .Or else ,it was disadvantage for cells growth .(5) Result indicated that the liquid medium of MS +1.0mg/L2,4-D+0.1mg/LKT+4% sucrose was the best in the suspension culture ; in the differentiation ,the solid culture medium of MS +1.0mg/L NAA+0.5mg/LKT was the best .(6) The cells of suspension were better material for chromosome doubling , considering result and economy , trifluralin would be treated as further study of chromosome doubling the Hybrid F1 between Leymus chinensis and Leymus cinereus .
