In this research, L.caise zhang DNA were peritoneal injection to mice then observed the macrophage phagocytosis rate,phagocytic index,macrophage C3b addiment and toxicity test. Then studied the immunizing synergism of L.casei Zhang DNA. Newcastle Disease and avian influenza respectively, the first immunitied at the 15th and 28th,booster after tow week, after the second immunity, sampled and detected antibody level and IgG content of the serum and lymphproliferation response by HA/HI,ELISA and MTT method respectively. The changes of IL-2 and IFN-γmRNA transcription levels of the AI tests were examined by semi-quantitative RT-PCR method at first week.The results of megalophage phagocytosis test indicated that the macrophage phagocytosis rate,phagocytic index and macrophage C3b addiment of the DNA group were significantly increased compared with the control group(p<0.05), Observing one week, we didn't find any adverse reaction of mice in toxicity tests.The result of the immunizing synergism of L.casei Zhang DNA indicated that the HI level,lymphproliferation response and IgG content of Immunity groups are higher than blank group super remarkably (p<0.05,p<0.01); Them of added adjuvant groups were higher than single immunizing antigen group (p<0.05,p<0.01); Added L.casei Zhang DNA group compare with only added and CFA group, that is not significant difference.Examined the changes of IL-2 and IFN-γmRNA transcription levels revealed that IL-2 and IFN-γcould be detected in chicken spleen lymphocytes at different degree level no matter in immuned group or control group. However the transcription levels of the two cytokines in immuned group were prominent higher than in control group and were significantly different between them (p<0.05,p<0.01). The transcription levels of the two cytokines in adding to adjuvant immuned group were prominent higher than in AI viccine immuned group and were significantly different between them (p<0.01); Added adjuvant group were not significant difference (p<0.05).
