In order to investage the effect of cysteamine on transformation of Peripheral blood T lymphocyte and the antioxidative capability of goats,the following experiments were carried out:ⅠEffect of cysteamine on transformation of peripheral blood T ly mphocyte in surgical goats.Part 1:Effeet of dietary nutrition and cysteamine in post-surgery periods on tran sformation of peripheral blood T lymphocyte in vive12 young goats experienced 3 periods:1) before surgery:On the basis of the bodyweight,the goats were divided into 2 groups:high level of nutrition(HL,n=6) and low level of nutrition(LL,n=6).Enough peanut stems and walter were provided,and 300g supplemental nutrition was added to HL,and 50g were added to LL.This period lasted 10 days.2) Period of surgery on rumen:after surgery,HL was divided into A(n=3)and B(n=3), and LL was divided into C(n=3) and D(n=3).From the first day after surgery,Cysteamine was added to A and C(45mg/kgBW·d),and supplemental nutrition was fed like before.3) Period of surgery on duodenum:14 days after period 2,surgery on duodenum was conducted and the mode of feed like period 2.The experiment finished on the 8th day. Peripheral blood was collected on the 8th day of period 1,the 4th,8th and 14th day of period 2,the1st,2nd,3rd,4th day and 8th day of period 3 respectively.Transformation of peripheral blood T lymphoeyte(TLC) was detected.Results showed supplemental nutrition exerted significant effete on TLC(p<0.05).TLC of group A and B decreased significantly on the 4th day after the surgery on tumen(P<0.05) and recoveried on the 8th day.But there was still predominant low TLC in group B(P<0.05).Fortunately,no significant difference between group A and before(p=0.12).After the surgery on duodenum,TLC of all groups decreased significantly(P<0.05).There was no difference between all groups on the 1st and 2nd day.On the 3rd,4thand 8th day,TLC of group A was significantly higher than others (P<0.05),and group D was the lowest(P<0.05) and there was no difference between B and C.So we concluded the following results:the supplemental nutrition can improve the function of lymphocyte in goats and cysteamine can dimish the celluar immunosuppress by preventing the decrease of TLC.Further,this improment wound be more better if we added cysteamine to supplemental nutrition.Part 2:Effect of cysteamine on transformation of peripheral blood T lymphocyte in vitroPeripheral blood lymphocyte were separated from 3 goats by density gradient centrifugation and cultured for 72h.Cysteamine was added to the lymphocyte in different concentration:0.25 mmol/l,0.5mmol/l,1.0 mmol/1,2.0 mmol/l,4.0 mmol/l respectively. Lymphocytes were collected and TLC was calculated.Compared with the control group, 0.25 mmol/l cysteamine has no effect on TLC,and 0.5 mmol/l and 1.0mmol/l cysteamine have positive effect(p<0.05)in dose effect.To contrary,TLC decreased in concentration 2.0 mmol/l and 4.0 mmol/l compared to the control.The result show that cysteamine has direct effect on lymphocytes and also has dose effect.Part 3:Effect of cysteamine and taurine on transformation of peripheral blood T lymphocyte under the condition of supplemental nutrition.7 healthy adult goats fixed permanent rurninal and duodenal fistulae were derided into group A(n=3) and group B(n=4),.And they experienced 4 periods1) Control period:beside peanut stems,each goat ate 200g supplemental nutrition everyday.2) period of experiment 1: supplemental nutrition plus eysteamine(45mg/kg.BW.d) were fed to group A,and supplemental nutrition plus taurine were fed to group B twice perday.3)recovery period: the forage fed like control period.4) period of experiment 2:the mode of feed was contrary to period 2.Each period lasted 14 days,and on the 12th day,TLC was tested.At the control and recovery period,there was no defference between A,and B(p=0.12,p=0.68), Significant defference emerged at period of experiment 1 and experiment 2(p<0.05).So we can say that addition of cysteamine and taurine can elevate TLC and relieve the cell immunosuppress after surgery.ⅡEffeet of cysteamine in post-surgery periods on antioxidation in goats6 young goats were derided into control group(n=3) and experiment group(n =3) according to the bodyweight.This experiment had 2 periods 1) supplemental nu trition period before surgery:This period lasted 10 days.Beside peanut stems,each g oat ate 300g supplementalnutririon everyday.2) Period of surgery on duodenum:after surgery,cysteamine(45mg/kg BW·d) was added to experiment group,and the supplem ental nutrition like before.This period lasted 14 days.Peripheral blood were collect ed on the 8th day of period 1 and the 1st,3rd day after surgery,yet on the 14th day after surgery the liver was gained to detect antioxidative index.The result was t hat on the 1st and 3rd day after surgery,the content of MDA elevated and the acti vity of SOD and GSH-Px decreased(P<0.05) significantly.There was no defference between two groups of MDA and SOD,but the extent of GSH-Px was lower in ex periment group than control(P<0.05).No difference occured between two groups in liver.It is obvious that surgery damaged the antioxidative capacity of goats and in this condition cysteamine has no effect to relief this damage.
