Adipocyte fatty acid binding protein (A-FABP) is a 15 kDa member of the intracellular fatty acid binding protein (FABP) family,which is known for the ability to bind fatty acids and related compounds (bile acids or retinoids) in an internal cavity. A-FABP is expressed in a differentiation-dependent fashion in adipocytes and is a critical gene in the regulation of the IMF in these cells of the animal.Therefore this experiment we selects a-FABP gene as the candidate gene of IMF,to study the SNPs of A-FABP gene and analyse the effect of polymorphism for meat quality in Songliao black pigs.Genetic polymorphism of Adipocyte fatty acid-binding protein gene's 5′UTR,intron 1 and exon2 in 46 pigs of Songliao black pig was detected by polymerase chain reaction- Single Strand Conformational Polymorphism. Polymorphism fragment of A-FABP gene was cloned and sequenced so as to ascertain variant location of polymorphism sites and mutational base type. The correlativity between these polymorphism sites and pig's meat quality traits was analysed with records of abatage determination,whose objective is that providing foundation of molecular biology for seed selection and selection about pig's meat quality traits,To study and analysis the gene variation in the molecular level of Songliao black pigs and to provide the theory basis of the relativity analysis with its economic character. According to detection outcome of PCR-SSCP,We didn't find the PCR-SSCP polymorphism sites in 5′UTR1,5′UTR3,intron1.1 and exon2 of A-FABP in Songliao black populations; we learned that polymorphism of Songliao black pig was detected in the 5′UTR2 and Intron1 sites of P5 primer. In 5′UTR2 site,there are two alleles -A,B and three genotypes-AA,AB and BB. The frequencies of A allele gene in Songliao black pig was 0.4348; In Intron1 sites of P5 primer,there are two alleles -E,F and three genotypes-EE,EF and FF. The frequencies of E allele gene in Songliao black pig was 0.6848.Throughχ~2 test,Songliao black pig hadn't achieve level of significance,I.e gene and genetype frequencies all achieved balanced state of Hardy -Weinberg(P>0.05). The results of cloning and sequencing indicated: 5′UTR2 polymorphism dued to base mutation of C→A in position 1321bp; A concatenate mutation of T→G in position 3440bp,A→G in position 3481bp and G→T in position 3514bp was detected in Intron1 sites of P5 primer,resulted in the polymorphism of intron 12.Correlativity between A-FABP gene's genetic polymorphism in Songliao black pig and meat quality traits was analysed with SPSS 10.0 software. Results indicated: in the Songliao black pig group different genetypes of 5′UTR2 site influenced IMF content extreme significantly (P<0.01) and influenced weeping loss,marbling significantly (P<0.05). IMF content: BB and AB was extreme significantly higher than AA (P<0.01),difference between BB and AB wasn't significant(P>0.05). Weeping loss: BB was significantly higher than AA and AB respectively (P<0.05),difference between AB and AA wasn't significant(P>0.05). Marbling: BB was significantly higher than AA and AB respectively (P<0.05),difference between AB and AA wasn't significant(P>0.05). Intron1 sites of P5 primer site polymorphism influenced marbling and IMF content extreme significantly (P<0.01). Marbling: FF was extreme significantly higher than EF and EE respectively (P<0.01). difference between EF and EE wasn't significant(P>0.05). IMF content: FF was extreme significantly higher than EF(P<0.01),and EF was extreme significantly higher than EE(P<0.01).A-FABP gene different genotypic combination's IMF content in Songliao black pig was as follows: Genotypic combination which IMF content was highest was''BB-FF",Least square mean of IMF content was 2.796. Thus it can be seen"BB-FF"was the best genotypic combination for improving the IMF of Songliao black pig. Research results cued that we could raise IMF content and improve meat quality by increase the frequency of best genotypic combination.
