Genetic polymorphism of Heart fatty acid-binding protein gene's 5'-upstream and intron 2 in 127 pigs including Songliao black pig,Junmuyihao white pig,PIC pig,Landrace pig,Large White pig,Duroc pig and Songliao-Boar crossing pig was detected by polymerase chain reaction- restriction fragment length polymorphism.Polymorphism fragment of H-FABP gene was cloned and sequenced so as to ascertain variant position of polymorphism locus and mutational base.The correlativity between polymorphism locus and meat quality traits in pig was analysed with records of abatage determination,whose objective is that providing foundation of molecular biology for seed selection and selection of meat quality traits in pig.According to detection outcome of PCR-RFLP, polymorphism of seven pig species was detected in the HinfI and HaeIII locus.The frequencies of H allele gene in Songliao black pig,Junmuyihao white pig,PIC pig,Landrace pig,Large White pig,Duroc pig and Songliao-Boar crossing pig were 0.7167,0.5600,0.6786,0.7500,0.7353,0.7333 and 0.8000 respectively; The frequencies of D allele gene were 0.3833,0.6400,0.4643,0.2813,0.6765,0.4000 and 0.3000 respectively. Other pig species showed polymorphism except that Songliao black pig,Landrace pig and Songliao-Boar crossing pig only showed AA in the MspI locus. Thefrequencies of A allele gene in Junmuyihao white pig,PIC pig,Large White pig and Duroc pig were 0.4800,0.3929,0.7647 and 0.6667 respectively.Seven groups including Songliao black pig didn't achieve level of significance throughχ2 test,I.E gene and genetype frequencies of seven groups all achieved balanced state of Hardy -Weinberg(P>0.05).The results of cloning and sequencing indicated: HinfI polymorphism of 5'-upstream dued to base mutation of T→C in position 1324; HaeIII polymorphism of intron 2 dued to base mutation of C→G in position 1811; base mutation of T→C in position 1489 resulted in MspI polymorphism of intron 2.Correlativity between H-FABP gene's genetic polymorphism in different pig groups and meat quality traits was analysed with SPSS 10.0 software.Results indicated: in the Songliao black pig group different genetypes of HinfI locus influenced marbling,weeping loss and IMF content extreme significantly (P < 0.01). Marbling:HH was extreme significantly higher than Hh and hh respectively; weeping loss: hh was extreme significantly higher than Hh and HH respectively;IMF content:HH was extreme significantly higher than Hh,and Hh was extreme significantly higher than hh. HaeIII locus polymorphism influenced marbling and ripe meat rate significantly (P<0.05),and influenced IMF content extreme significantly (P<0.01). Marbling: dd and Dd were significantly higher than DD respectively; ripe meat rate: Dd was higher than DD significantly,difference between Dd and dd wasn't significant, difference between dd and DD wasn't also significant;IMF content: dd and Dd were extreme significantly higher than DD respectively.In the PIC pig group different genetypes of HinfI locus influenced IMF content extreme significantly (P<0.01), Hh was extreme significantlyhigher than HH and hh respectively. HaeIII locus polymorphism influenced IMF content significantly (P<0.05), Dd and dd were significantly higher than DD respectively. MspI locus polymorphism influenced IMF content extreme significantly (P<0.01), aa was extreme significantly higher than Aa and AA respectively.In the Landrace pig group HinfI locus polymorphism influenced carnation and weeping loss significantly (P<0.05). Carnation: Hh was higher than HH significantly,difference between Hh and hh wasn't significant, difference between hh and HH wasn't also significant; weeping loss: hh and HH was significantly higher than Hh respectively. HaeIII locus polymorphism influenced IMF content extreme significantly (P<0.01), dd was extreme significantly higher than Dd and DD respectively.In the Large White pig group HinfI locus polymorphism influenced IMF content significantly (P<0.05). HH and Hh were significantly higher than hh respectively. HaeIII locus polymorphism influenced shearing force and IMF content extreme significantly (P<0.01). Shearing force: Dd was extreme significantly higher than DD and dd respectively;IMF content: dd was extreme significantly higher than Dd and DD respectively. MspI locus polymorphism influenced weeping loss significantly (P<0.05), AA was higher than aa significantly,difference between AA and Aa wasn't significant, difference between Aa and aa wasn't also significant.In the Duroc pig group HinfI locus polymorphism influenced water depletion rate extreme significantly (P < 0.01), HH was extreme significantly higher than Hh and hh respectively. HaeIII locus polymorphism influenced ripe meat rate and IMF content extreme significantly (P<0.01). Ripe meat rate: DD was extreme significantlyhigher than Dd,and Dd was extreme significantly higher than dd;IMF content: dd was extreme significantly higher than Dd,and Dd was extreme significantly higher than DD. MspI locus polymorphism influenced water depletion rate extreme significantly (P<0.01),AA was higher than aa extreme significantly,difference between AA and Aa wasn't significant, difference between Aa and aa wasn't also significant.In the Songliao-Boar crossing pig group HinfI locus polymorphism influenced weeping loss extreme significantly (P<0.01). hh was extreme significantly higher than Hh,and Hh was extreme significantly higher than HH. HaeIII locus polymorphism influenced IMF content extreme significantly (P<0.01), dd was extreme significantly higher than Dd and DD respectively.H-FABP gene different genotypic combination's IMF content in different pig species was as follows: genotypic combination whose IMF content was highest was"dd-HH","dd-HH-AA","DD-HH","Dd-HH-aa","dd-HH-aa"and"Dd-HH"respectively in Songliao black pig,Junmuyihao white pig,PIC pig,Landrace pig,Large White pig,Duroc pig and Songliao-Boar crossing pig.Least square mean of IMF content was 2.784,1.880,2.340,1.430,3.510 and 2.400 respectively.Research results cued that IMF content could be raised and meat quality could be improved by increase the frequency of best genotypic combination.
