This paper investigated different detection methods of Eperthrozoonsis, and the establishment of animal model and study of the pathogenicity of this disease.1. The ultramicrostructure of Eperythrozoon suis was observed through light microscope (blood smear or M-Wright's staining method) and transmission electron microscope. The structure of EH could be clealy observed both in blood smear and using M-Wright's staining method, but there may be some false positive when using these two methods.So PCR is needed for auxiliary diagnosis. Electron microscopy is useful when observing the ultramicrostructure of EH, but it's complicated, so it's not suitable for clinical use.According to published genome sequence of Eperythrozoon suis, a set of primers were designed and a 801bp PCR product was obtained. The PCR method was highly specific and sensitive and could be used for EH epidemiologic survey. At the same time, a specific and reliable real time PCR method was developed in this study.It can be used for further study.2. In this report, the whole blood of a pig suffering from Eperythrozoosis and purified Eperythrozoon suis were used as sources of infection.SPF KM mouse were used as experimental animal. Using microscopic observation of erythrocytes, PCR analysis and transmission electron microscope (EM) detection methods, the mouse model was characterized.The results showed that using purified Eperythrozoon suis as source of infection was better than using the whole blood of a pig suffering from Eperythrozoosis.The model could be used for the further study.3. In this report, the haematological and immunologic characteristics were studied on the basis of mouse model we established. The results showed that total red cell coμnt, erythrocrit, and haemoglobin levels were remarkably lower in experimentally-infected mice, and the rosette formation rate of red blood cell immune complexes were remarkably increased in experimental group.This suggested that EH infection may led to the impairment of erythrocyte immune system,but it couldn't arouse a strong specidic immune response.In this paper, a detection method including microscope, transmission electron microscope and PCR was established. A mouse model of Eperythrozoon infection was established to confirm inter-species infection, and the haematological and immunologic characteristics were studied.The pathopoiesis mechanism was discussed according to the result. This study provided a theory and technique reserve for EH epidemic outbreak, and it's significative for formulating the integrate control program of EH.
