In order to get better understanding the impacts of radiation on Gladiolus generation M2 quality and biological characteristics, this experiment planted three varieties of Gladiolus generation M2 with five-dose-radiation named " Ovutie " " Advance red " and " Spic and span " on the rest fields, observed and detected their morphological indicators, and then compared the results with the generation M1, the results showed that the morphological indicators of Gladiolus generation M2 inherited the variations characteristics of generation M1, and each indicator decreased with the increase of radiation dose. Compared with the generation M1, each indicator of generation M2 of low-dose radiation did not show higher than controls, except in number of the ball diameter and the amount of new balls, this indicated that the promotion impacts of low-dose-radiation on the growth of Gladiolus only expressed on the contemporary radiation, this radiation has little effect on its future generations. In 13 individual plants of Generation M1 of fancy point's variation, 10 individual plants of generation M2 maintained their variation traits, 3 individual plants lost their variation characteristics.We also found new variation in generation M2 which did not emerged in generation M1. An individual plant with pink petal edge which emerged in " Ovutie " variety has high ornamental. An individual plant with stamen petalody emerged in " Advance red " variety. In"Spic and span"variety, an individual plant with amphichrome emerged; and another individual plant with color variation emerged, its color was yellow and green which also has high ornamental.In order to facilitate the post-radiation Gladiolus to be a crossbreeding material, the experiment of detected the pollen activity of the varieties of " Ovutie " and " Spic and span " with different radiation dose, then, we can get suitable for the best radiation dose for crossbreeding. The results showed that the group of 25 GY has the strongest pollen activity in both groups; the control group has less pollen activity, the 75 GY is the weakest group. In all dose groups, 25 GY radiation dose and 50 GY were suitable to use as hybrid breeding materials.This experiment detected RAPD of 11 mutants which have special variation in the M2 generation, its aim is to take use similar mutation of the individual to find similar specific fragment in DNA group of Gladiolus, and then determine the corresponding relations between the fragment and characters. The results showed that the different specificity of band emerged compared with controlled plant in each mutation, but similar characteristics of the specific bands were not found in a similar mutation. There could be following reasons through analysis: 1. RAPD primers were relatively small quantity, and it failed to amplify to the gene which can link its character. 2. The amount of Mutants is relatively small, there were only two individual plants with a similar traits of mutant emerged in the largest group, It is difficult to detect the specificity of this band under such small quantity. 3. Due to the equipment, the limitation of understanding of RAPD reaction system, also with the limitation of the operation proficiency and the time constraints, test was failed to achieve the best results, with many bands were not amplified clearly enough, some bands were not even amplified to determined. We should sum up experience in the future, improve the experimental conditions, and finally get the perfect test results.
