In order to study the effects of the physiological change in the K male-sterile line and the YS temperature sensitive male-sterile line pollend evelopment process on the fertility of wheat, Investigated the physiological mechanism of pollen abortion and rule of changeover of sterility with comparing RNase, soluble protein, GSH-px,MDA content change of the K male-sterile line and the YS temperature sensitive male-sterile line Sampling from different environment,investigating physiological mechanism of pollen abortion and fertility alteration of rule.This provide theoretical basis and technical support for effectively cultivating and using cytoplasm male-sterile line.The results of the present study are valuable in increasing yield and improving quaility by genetic and molecular biology. The research results are the following:1.RNase activity of 3314A and 732A (K male-sterile line) increased before dinucleate, significantly higher than the maintainer line, respectively is 1.36 times and 1.30 times of the maintainer line. Soluble protein in the pollen of male-sterile decreased between uninucleate and dinucleate, soluble protein content is 1.36 times and 1.30 times of the maintainer line significantly lower than the maintainer line. RNase activity increased abnormaly lead to decreasing of soluble protein content and physiological metabolic disorder,which may cause pollen abortion,so dinucleate is key period of the pollen abortion possibly;RNase of 732A and 3314A was significantly different from the maintainer line, RNase activity of the leaves was lower than the pollen. Soluble protein content of 3314A and 732A (K male-sterile line)and the maintainer line pollens changed more obviously than leaves in whole growth period, therefor,the effects of RNase activity and soluble protein of the pollens on wheat fertility were strong. RNase and soluble protein of A3017(YS temperature sensitive male-sterile line) under fertile condition was significantly different from under sterile condition,and obviously different during fertility alteration, which may be related to fertility alteration.2. GSH-px activity of 3314A leaves and pollens and 732A leaves and pollens decreased in the pollen development period, and higher than maintaitwo nuclei are higher than the maintainer before dinucleate, lower than the maintainer after dinucleate, but MDA content increased,higher than the maintainer. GSH-px activity of A3017 under sterile condition lower than under fertile condition, which indicated that GSH-px activity was related to pollen abortion and fertility alteration of temperature sensitive male-sterile line. GSH-px of male-sterile line in pollens and leaves developmental stages, it did not eliminate O2-and oters timely, caused O2- and others accumulation, the lipid peroxidization inmembrane was intensified, causes the accumulation of MDA , the damage of the pollen membrane system, influenced the development of pollen finally.3. A3017 and 732A are near-isogenic lines (NILs),the temperature sensitive characters were different, but change trend and value of RNase activity,soluble protein content, GSH-px activity,MDA content were similar,however, RNase activity,soluble protein content, GSH-px activity,MDA content of A3017, 732A were different from 3314A(1B/1R type)significantly. This possibly is because 1RS chromosome of 1B/1R the type has substituted the 1BS.4.RNase and GSH-px activity,the protein content and MDA content of 732A, 3314A and the maintainer were not influnced by temperature.But RNase and GSH-px activity,the protein content and MDA content of A3017 were remarkably different under the different temperature. The result indicated that RNase and GSH-px activity,the protein content and MDA content of pollen changed remarkably during fertility alteration, were related with fertility alteration of temperature sensitive male-sterile line .
