Research On Active Proteins And Atpase Activities In The Fertility Sensitive Period Of Wheat Male Sterile Lines

To research the correlativity between ATPase activity in plasma membrane of spikes, ATPase activity in tonoplast of spikes , difference of protein,ultrastructure of anthers and male sterility ,ATPase activity of male sterile wheat lines and their maintainers was measured, active protein was analyzed by native-PAGE and the ultrastructures were conducted on pollen development by transmission electron microscope. The result of research are as follows:There are high ATPase activity level in non-1B/1R type male sterile line 732A and YS type male sterile line 3017A,while low ATPase activity level in maintain line 732B and restorer line 160 by research of ATPase activity in plasma membrane of spikes at their uninucleate stages. At their binucleate stages, result is 3017A with a higher ATPase activity level, while other male sterile lines and their maintainers and restorer line with lower ATPase activity level. Whereas variation trend of ATPase activity in plasma membrane is much similar in 1B/1R type male sterile line 3314A and its maintainer 3314B, with high level at their uninucleate stages, low level at their binucleate stages. So it suggest that there is a correlativity between ATPase activity in plasma membrane and sterility of 732A and 3017A,but this correlativity is not exist in 3314A.Then ATPase activities were measured in tonoplast of spikes respectively. The results reflected that there are higher ATPase activity levels in tonoplast of the spikes of sterile lines compared with their maintainers at their uninucleate stages, and become a low level in all materials without 3017A at their binucleate stages. So there is a correlativity between ATPase activity in tonoplast and sterility of these several male sterile lines and male sterile lines present a high ATPase activity level. Uninucleate stage is fertility sensitive period of 732A and 3314A, while uninucleate stage and binucleate stage are both Fertility Sensitive Periods of 3017A.Research on Active protein in flags of male sterile wheat lines and their maintainers by Native-PAGE reflect that, special protein was found in male sterile line 3314A and maintainers 732B at binucleate and trinucleate stages, whereas it was not found at uninucleate stage, yet not found in other materials at their uninucleate, binucleate and trinucleate stages. This special protein is both found in sterile line and maintainers reflect that it is not a relationship with fertility development. So we haven't found special protein affect fertility development.Research on Active protein in spikes of male sterile wheat lines and their maintainers by native-PAGE reflect that, more special proteins were found in non-1B/1R type male sterile line 732A and 1B/1R type male sterile line 3314A. These special proteins possibly lead to male sterility. There is still a special protein with low molecular weight in all materials at their binucleate stages but not exist at their uninucleate stages .This protein may be necessary in development of spikes but not necessary in fertility development. The result also reflect that uninucleate stage is fertility sensitive period of 732A and 3314A,and maybe special protein expressed in male sterile line lead to abortion.The ultrastructures of 3314A and its maintainers 3314B, 732A and 732B were conducted on anther development at their uninucleate, binucleate and trinucleate stages by transmission electron microscope. The results show that there is a thin cytoplasm in 3314A at its uninucleate stage and no ubisch body at its binucleate stage , while amount of ubisch body in its maintainer 3314B at its binucleate stage. At trinucleate stage, tapetum clear up and pollen grain become crimplein in 3314A, whereas tapetum still exist and pollen grain is in spherical shape in 3314B. The nucleus was shouldered to side, material distribution and no tapetum found in non-1B/1R male sterile line 732A at its uninucleate stage, whereas the pollen was surrounded by tapetum in 732B. At trinucleate stage,there are many large starch grains in pollen of 732A, and tapetum clear up ,pollen grain is crimplein ,while there are many small starch grains in pollen of 732B and tapetum still exist, in spherical shape.
