The experiment was conducted by comparative analysis of pollen germination rate, soluble sugar, starch, protein and proline content, POD isozyme and esterase isozyme of anther as well as the POD isozyme of leaves, between the male-sterile lines and fertile lines, under the natural environment temperature and artificially controlling temperature. The study of this thesis was to provide theoretical basis on physiological and biochemical for male sterile mechanism of thermo-sensitive male sterile line tomato and to make a better use of it.The results included as follow:1. Pollen germination rate of "T-4" was below 30% from June 26th to September 2th, but it was more than 30% from November 3th to December 8 th. That fertility of "T-4" was related to night temperature was speculated.2. Low night temperature had some effect on fertility restoration. It played a key role in fertility restoration of "T-4" when it was at pre-flowering 11-14d.3. The critical temperature of fertility alteration of "T-4" were 13.93℃under low night temperature conditions. The critical temperature of fertility restoration of "T-4" was 15.59℃.4. Pollen germination, seeds number,the proline, starch, soluble sugar and protein content of "T-4" under the two conditions (at day 28℃/night 24℃and day 28℃/night 12℃) were respectively significantly lower than those which were under the condition of day 28℃/night 18℃. There were no significant differences between "T-4" at day 28℃/ night 18℃and fertile line "First". It illustrated that "T-4" restored fertility partly. 5. The POD isoenzyme band numbers of the leaves and anther, the EST isoenzyme band numbers of the anther in "T-4" in fertile conditions (at day 28℃/night 18℃) were much more than those which were in sterile conditions (at day 28℃/night 24℃and day 28℃/night 12℃). So POD enzymatic activity of the leaves and anther in male-sterile line was higher than the sterile line.6. "T-4", which were in the four periods of flower bud differentiation period (2th leaf stage), stamen differentiation period (6th leaf stage), pistil differentiation period (7th leaf stage) and bud period (7-8 d before blossom), were treated with 3d,6d and 9d under different temperature treatments (day/night 28/12,28/18 and 18/24℃).The results showed: pollen germination rate of "T-4" in flower bud differentiation period, pistil differentiation period and bud period was respectively below 30%.There were no significant differences between "T-4" and its fertile line "First". However, pollen germination rate of "T-4", which were in stamen differentiation period,was up to 33.78% when treated with 9d at day 28℃/night 18℃. Otherwise, when treated with 3d and 6d it was below 30%. So it stamen differentiation period that was the sensitive period of "T-4".7. Pollen germination rate, soluble sugar, starch, protein and praline content of "T-4" which were treated respectively with 3d,6d and 9d in flower bud differentiation period, pistil differentiation period and bud period, had significant differences with its fertile line. pollen germination rate of "T-4". But those which were treated with 9d in stamen differentiation period has no significant differences with its fertile line; on the contrary, when they were treated with 3d and 6d. So stamen differentiation period was the sensitive period for "T-4".
