A referencing flock from five groups in Macaca mulatta lasiotis was utilized to investigate the genetic diversity by polymerase chain reaction(PCR) and Polyacrylamide gel electrophoresis by followed silver staining. The genetic heterozygosity, Polymorphism information content(PIC), effective numbers of allele, genetic distance among Macaca mulatta groups were computed and dendrogram were constructed to evaluate the genetic variability within populations and genetic relationships among the groups.1) 25 microsatellite loci are used in this experiment and 8 microsatellite loci amplified well were used in the 111 samples of the 5 populations. 109 allelic were got in all the loci and the average number was 13.62 in one locus, the average effective numbers of allele was 6.52 in one locus and explain that the 8 microsatellite loci in this experiment were in high polymorphism.2) The Hardy-weinberg equilibrium analysis 5 populations revealed that expect MML10S6F in the Bazhong population and D17S1300 in Jiulong population, the other loci all showed a remarkable genetic disequilibrium in this two populations. In the other populations the equilibrium loci and the disequilibrium loci were in equal to each other. as a whole, the Macaca mulatta lasiotis 3 loci showed a remarkable genetic equilibrium and the other 5 loci showed a remarkable genetic disequilibrium. Analysis its causes, may be due to the Stocks of sub-structure, Inbreeding and the null alleles.3) The 8 microsatellite loci showed high PIC value and polymorphism, the average PIC value was 0.89, the observed heterozygosity was 0.64.4) Nei's genetic distance were estimated and used to conduct UPGMA and N-J dendrograms. The UPGMA and N-J dendrograms based on the Nei'genetic distance showed a short genetic distance between Xiaojin and Heishui,Hanyuan and Jiulong, and then had a longer distance with Bazhong population. The clustering order conformed to the geographical distribution of five populations. Hanyuan and Jiulong located in the west of Sichuan, Bazhong located in the east of Sichuan, Xiaojin and heishui located in the northwest of Sichuan. The genetic distance between the 5 populations is accurate.
