The study was mainly to probe an efficient way through tissue and rapid propagation of Pistacia weinmannfolia.The stems with shoot,the leaves,the leafaxles of young branches were taken as explants.Orthogonal experimental design and analysis of variance were used to establish the technique system of tissue culture,the affected factors of propagation,existing problems in this technique and the corresponding solutions were also described,which provide the theoretical base and technical measures for industrial producing of plantlet.The study of tissue culture technique and the better medium for various stages were as follows:①April was the best season to draw materials as stems,leafaxle,in which tlae germination rate were 94.00%and 90.00%by stem and leafaxle respectively,the contaminated rate only were 21.67%and 20.33%by stem and leafaxle respectively.The leaves nearly did not germinate,but its contaminated rate was lowest in April,in which the germination rate was 18.00%,the contaminated rate was 3.00%.②During the trial:7 minutes was good to treat the stem by HgCl2(0.1%),in which the germination rate was 85.00%,the contaminated rate was only 25.00%.5 minutes was good to treat the leaves and leafaxle,in which the germination rate were 2.00%and 80.00%,the contaminated rate were 29.33%and 30.00%.③Initial culture:Initial medium of stem:MS+6-BA0.5mg·L-1+2,4-D0.01 mg·L-1,in which the germination rate was 99.33%,the callus induction rate was 86.67%,and the shoot forming index was 1.8;Initial medium of leafaxle:MS+6-BA 0.5mg·L-1+NAA 0.1 mg·L-1+2,4-D 0.05 mg·L-1,in whichthe callus induction rate was 85.00%. ④Proliferating and callus of stems differentiating cultivation:The optional proliferating buds culture medium stems and shoots was:MS+6-BA 0.5mg·L-1+2, 4-D 0.1mg·L-1+NAA0.1mg·L-1,in which the multiplication rate was 4.1 and the multiplication of efficiency seedings was 3.5.Highly proliferating callus was obtained following cultivation on MS+6-BA 1.0 mg·L-1+NAA 0.1mg·L-1+2,4-D1.0mg·L-1.The proliferating rates could be up to 83.53%.The best culture medium for callus of stems differentiating was MS+6-BA 1.0mg·L-1+ NAA 0.1mg·L-1.The regeneration rate was 74.51%.⑤Rooting culture:1/2MS+ NAA 0.1 mg·L-1+IBA 1.0 mg·L-1+AC0.3%,in which the rate of rooting was 82.5%,the quantity of roots was 4.9,and the length was 3.3cm.⑥Forging seeding transplanting:The transplant medium in 25%of vermiculite, 25%of perlite and 50%of riversand was the optimal for Pistacia weinmannfolia, which the rate of survival was 68.00%.
