Studies On Tissue Culture And Browning Of Pistacia Vera. L.

Pistacia.vera.L.is one of the nuts and has high nutritional and economic value,,but it is so lack in china,now the main way to get them is importion from Iron and America.So heavy cash is running away.The purpose of the research is to do some work with the method of tissue culture to find out the best condition of its quick propagation.Tissue culture is important in research of plant.Sterm is selected as explant in this article,in order to establish the regeneration systems of Pistacia. The best combination of hormones and the characters in physiology are studied in this research. Conclusions are as follows:(1) Young stem is good material as explant in tissue culture of Pistacia.vera.L.And the rate of split is up to 80%.The leave does not suit for tissue culture because of high rate of browning.(2)Adding some hormones can lead to the chage of hormones inside plants.In the early days of split and propagation , hormones inside have increased;and with the days go on ,then to discrease..(3) Comparing with several factors that affecting the browing reaction and comtamination,the suitable medium and combination is: DKW(Modified)+6-BA4.0mg/L+IBA0.05mg/L+NAA0.02mg/L.(4) The plan 1/2DKW+6-BA3.0～4.0mg/L+IBA0.02mg/L+NAA0.05 mg/L is used in tissue can get high rate of propagation.(5) The consistency of K~+ affects regeneration and browning reaction.(6) The browning connects to polyphenols directly,the higher of weight the heavier of browning; the proliferation times and the days are more,the contents of polyphenols will come up,then make browning deeply.(7) Through the research on chages of PHysiology during different organs ,the same organ indifferent seasons and the tissue plant ,The activity of PPO does not fluctuate largely,and not affect the browning reaction,on the contrary,the browning is affected by POD,SOD,PAL.
